ObjectiveChina Minority Medicine is part of Chinese Medicine which created by the55minotities. As one of those minotities,Tujia also has a long history of application of ethnic drugs, and the traiditional culature needs to be inherited and developed.This paper fairly comprehensively investigated the previous studies of this plant called Pittosporum Illicioides.This experiment aims to research and establish the fingerprint for Pittosporum illicioides by HPLC coupled with the UV detection,tracking the two flavonoid glycosides respectively named Tamarix prime-3-O-rutin glucosidase and Rutin.By analysing the date on obtained chromatograms to explore the influence on the content of the two flavonoid glycosides in the leaves of pittosporum illicioides collected at different time.Also the content determination of the two flavonoid glycosides has been done.,which provide the experimental date for the standardization of collection and the initial processing course of the leaves of pittosporum illicioides. The volatile components in the leaves of pittosporum illicioides were analyzed by GC-MS which lay a new foundation for further research of this plant.MethodsChoose10bacth of leaves from Pittosporum illicioides,after some pretreatment including cutting up the leaves and ultrasonic extraction,then determine the content of Tamarix prime-3-O-rutin glucosidase and Rutin by HPLC. The analysis was performed on chromatographic column Agela Technologies Promosil C08(4.6x200mm,5um) with gradient mobile phase consisted of methanol (A)-0.4%phosphoric acid water(B).The UV detection wavelength was254nm,the column temperature was40℃with the flow rate of1.0ml.min-1and the analysis time was60min.The fingerprint date was processed by the method of similarity.The volatile components in the leaves were analyzed using GC-MS combines with computer search and their relative contents were identified by peak area normalization method.ResultsThe results showed that there is a big difference in content of Tamarix prime-3-O-rutin glucosidase and Rutin in leaves from Pittosporum illicioides whose relative contents are0.04-0.07%and0.05-0.52%,and the third batch of this plant has the maximum content, respectively are0.07%and0.52%.Choose10bacth of leaves from Pittosporum illicioides,the date analysis showed that there was some difference in similarities.3bacth of these samples were obviously different from the left,whose relative similarity date were under0.9and between0.930-0.980.28kinds of compounds in the essential oils from the leaves have been identified,The main components in the kinds of essential oils includesn-und ecane(56.22%),caryophyllene(9.47%),germacrene-B(7.35%),caryophyllene-oxide(6.32%).Conclusions It indicated that there is some difference in the content of the two flavonoid glycosides., and reveals there is a small difference between the left7batch while a big difference existed between the3batch and the7batch. The method of the fingerprint established for Pittosporum illicioides by HPLC coupled with the UV detection has provided some model for the overall qualitative and quantitative research.There are28kinds of constituents of essential oil which lay some foundation of the research of therapeutic effects of the substance in the leaves of pittosporum illicioides.There are two innovation points in this paper.One is that the volatile components in the leaves of pittosporum illicioides were analyzed by GC-MS and28kinds of constituents have been identified for the first time which lay a new foundation for further research of this plant. The other one is that the method of the fingerprint by HPLC coupled with the UV detection for Pittosporum illicioides has been established for the first time which provide some model for the overall qualitative and quantitative research. So these results has afforded us a scientific basis of comprehensive utilization of the leaves of pittosporum illicioides.
