| Objective:Using the surface enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS) technology to detect and find out the serum differentially expressed proteins of the blood stasis syndrome of IgA nephropathy before and after taking hirudo(Huoxuetongmai capsules),which based on:1.Comparing IgA nephropathy group with the control group;2. Comparing the blood stasis syndrome of IgA nephropathy and non-blood stasis syndrome of IgA nephropathy.Testifiesing the serum specific expression protein,and provide an objective basis for the treatment of the blood stasis syndrome of IgA nephropathy at the protein level.Methods:Reference to the classification criteria of TCM syndrome for chronic primary glomerular disease which is adopted by second national nephropathy thematic colloquium,as well as the international blood stasis diagnostic criteria, formulated by international conference on research stasis syndrome in1988,with a combination of laboratory coagulation indicators.We classfied outpatient and ward inpatients kidney patients from October2011to February2013in Fuzhou General Hospital of Nephrology into different TCM syndrome group.We give stasis syndrome of IgA nephropathy patients medicine,who consent to taking hirudo(Huoxuetongmai capsule,5gram,3times a day) for4weeks.Proteomics methods (SELDI-TOF-MS technology and metal ion chelating protein chip) were applied to explored the serum protein fingerprints of each group(including14cases of blood stasis syndrome of IgA nephropathy and10cases medication group,16cases of non-blood stasis syndrome,14serum samples of the non-IgAN nephropathy patients and15blood samples of the control group), screening for the differentially expressed proteins in serum, in order to verify improvement of the hirudo to the blood stasis syndrome of IgA nephropathy based on experimental index:1st,the IgA nephropathy group and the control group;2nd,the blood stasis syndrome group and non-blood stasis syndrome group in the IgA nephropathy group,and the special expressed proteins for the blood stasis syndrome were identified by Biomarker Patterns Software to create the syndrome decision model;3rd, the blood stasis syndrome of IgA nephropathy before and after taking hirudo(Huoxuetongmai capsules).Result:1st,According to the comparison of the plasma protein fingerprint datas, it was found that30protein peaks were significantly different between the IgA nephropathy patients and the control group (P<0.05).2nd, after comparing the plasma protein fingerprint datas of IgAN between blood stasis syndrome group and non-blood stasis syndrome group, we found that42protein peaks were significantly different (P<0.05),the protein peaks were analysised by BPS software and found that7protein peaks were the best markers for the blood stasis syndrome(M/Z were1092.71ã€1972.32ã€2687.74ã€3196.19ã€3249.02ã€8567.20ã€8713.48).3rd, we also compared the plasma protein fingerprint datas of the blood stasis syndrome of IgA nephropathy before and after taking hirudo(Huoxuetongmai capsules), we found that15protein peaks were significantly different(P<0.05).4th,on the basis of the above results,we found that the protein of M/Z3249.02ã€8567.20and8713.48are likely to be the specific proteins of the blood stasis syndrome of IgA nephropathy,combined with manual screening and group analysis of variance,the protein of M/Z8713.48are significant findings.Conclusion:1.Combining software analysis with manual screening,we found that:1st,the protein of M/Z8713.48has a high expression in IgAN group with blood stasis syndrome.2nd, hirudo can alleviate the symptoms of the blood stasis syndrome of nephropathy though interfering the expression of the protein of M/Z8713.48. |
