Study Of Purification, Antitumour And Safty Of Ussurin From Gloydius Ussuriensis

Disintegrin is composed of49-84amino acid,which is a peptide rich in Cyt.It hasa RGD motif surruongded by–S-S-bridge. It has potent to inhabit aggregation ofplatelet,maybe a piece of hydrolysation of pro-metalproteinase.Snake disintegrin is distributed grop according to the amino acid longs:â‘ shortstring disintegrin,compoed of49-51amino acid.â‘¡medium string disintegrin,composed of70amino acid including12Cyt.â‘¢long string disintagrin,composed of84amino acid including14Cyt.â‘£two string disintegrin, each string is like shortstring.In our test,we used venom of Gloydius ussuriensis and get a novel peptidecomponent,named ussurin by using chromatograph of CM-Sepharose FF,DEAE-Sepharose FF, Sephadex G-100and Sephadex G-75.Ussurin is a peptide of7.4kD and a single band in reduction akalinSDS-PAGE.Using ussurin to test inhibition of aggregation of platelet,anti-tumorability and safty, we found that ussurin has the potent of inhibition of plateletaggregation.Its anti-tumor activity of IC₅₀to MDA-MB-231、A549、Hela is about6.1μg/ml、6.1μg/ml and6.9μg/ml. The toxicity of LD₅₀of ussurin in the mice is1481mg/kg and is1383mg/kg in the rat. The toxicity of ussurin is low and has noimmunogenicity hemolytic activity.Ussurin is great value in the future medical use.
