Construction Of Cell-based Models For Screening Sirt1Activator And Preliminary Application

Natural medicine has a long history in China. With its wide range of sources andmild action, natural medicine has been widely used. With the development ofextraction and purification technology natural medicine in recent years, scientistshave get a lot of monomer compounds which have good effect. It is reported sirt1gene plays an important role in some of the energy metabolic diseases, such asdiabetes, liver disease. Therefore regarding sirt1gene as a target for drug screeningmay help us to get some of the drugs which have a therapeutic effect for such diseases.The water extract of natural medicines is the most frequently used ingredient of naturemedicinal, therefore our experiment uses the water extract of natural drugs as a drugscreening library.With the deep study of luciferase in recent years, we find that it not only can beused to mark cells or molecules for cellular tracking and body imaging but also can beapplied to the drug high throughput screening. Through dual luciferase reporter genesystem, we can quickly filter out the desired molecular from small moleculecompound libraries. Due to its high sensitivity, specificity, easiness to operate andhigh-throughput, dual luciferase reporter gene system has been widely used. Thisexperiment makes sirt1promoter be cloned into the reporter gene luciferase promoterregion by dual luciferase reporter gene method. The method of detecting luciferaseactivity is tested by transient transfection cells administered after stimulation. Toestablish a sirt1activator cell screening model, the experiment regards water extractsof selected natural medicine as a drug screening library to conduct sirt1activator screening, expecting to obtain the number of sirt1activators and to find a new methodof treatment of such energy metabolism diseases.
