This paper belongs to the national basic research program (973Program) drug related research on the basis of theory--Chinese medicine system resistant material basis and specific effect on research, mainly carried out effective parts of Styrax GC-MS analysis and pharmacokinetics study.Methods:①This paper according to the China pharmacopoeia (2010edition) and Styrax the quality testing.②Applying the GC-MS technology, the author analyzes the compounds among solvent extraction (by petroleum ether and ethylether.), extraction of volatile oils through steam distillation and extraction of volatile components through HS-SPME.③This paper established the cinnamic acid as an index of Styrax pharmacokinetic study method by HPLC.④The content of Cinnamic acid in rat plasma, brain, liver, heart, spleen, lung, kidney were determined.Results:①According to the "Chinese Pharmacopoeia"(2010Edition) Styrax under were quality testing, test Styrax medicine Pharmacopoeia standard.②By GC-MS technology,27compounds were identified through the extraction of the solvent of Styrax, including23compounds in the part of petroleum ether and10compounds in the part of ethylether.42compounds were identified during the volatile oils extracted by steam distillation.28compounds were identified through HS-SPME. The components obtained through three methods all include10chemical constitutions such as a-pinene, Camphene, Benzenepropanoic acid,(-)-Verbenone and4,6,6-Trimethyl-bicyclo[3.1.1]Hept-3-En-2-One.14components obtained through solvent extraction are same as those obtained through steam distillation.24components obtained through steam distillation extraction are same as those obtained by HS-SPME.③he establishment and the determination of cinnamic acid as indicators of Styrax pharmacokinetic study method and pharmacokinetic parameters:plasma protein precipitation with methanol treated by HPLC analysis, in the plasma of cinnamic acid in0.125~64μg·mL-1range of cinnamic acid peak area (A) with good linear relationship with the concentration of X (r=0.99995); Precision of the test in low, medium and high concentration of three kinds of quality, days after RSD precision than RSD5.66%;3.76%and4.16%,3.49%,1.08%and3.71%; stability test in low, medium and high concentration of three kinds of quality RSD respectively0.72%,0.78%and1.75%; extraction recovery of low concentration of the average extraction rate of recovery is74.62%, RSD is3.17%; the average concentration of extraction rate of recovery is81.60%, RSD is2.45%; high concentration average extraction rate of recovery is81.91%, RSD is2.92%.9rats were given intragastrically Styrax medicine, establishment of cinnamic acid plasma concentration-time curve, calculated the corresponding drug kinetics parameters. The major pharmacokinetic parameters,female:Cmax=19.0228μg·mL-1;Tmax=24.1367min;AUC=2689.593μg·m L-1;male:Cmax=10.6980μg·mL-1;Tmax=28.6939min;AUC=1303.6717μg·mL-Pharmacokinetic parameters showed in Styrax cinnamic acid in rats in vivo drug process in line with the two compartment model.④The Styrax in rat lung, liver, spleen, kidney, heart, brain tissue are the distribution, the average content in the order of0.8048μg·g-1,0.6850μg·g-1,0.6513μg·g-1,0.3924μg·g-1,0.3798μg·g-1,0.1229μg·g-1.Conclusion:This study conducted a Styrax effectively part of the GC-MS study, analysis of the different extraction methods of volatile components of the similarities and differences, for further research and fragrant incense play to begin to Styrax the effectiveness of the drug research lays the foundation, to further confirm the Styrax play aroma resuscitation of pharmacodynamic material basis of; of cinnamic acid in storax in rats in vivo pharmacokinetic process and cinnamic acid distribution in rat tissues situation, explore the in vivo process, in order to explain the theory of traditional Chinese medicine and traditional Chinese medicine clinical medicine foundation.
