Objective:MNU of rats induced by apoptosis photoreceptor cells model as the research object, this paper discusses the acupuncture intervention photoreceptor cells apoptosis related mechanism,for prevention and control of acupuncture photoreceptor cells of the retina apoptosis eye disease provide certain experiment basis.Methods:The MNU SD rats induced apoptosis photoreceptor cells animal model, randomly divided into:acupuncture group, vitamin A group, model group, and normal control group, continuous treatment after14days, the mfERG, HE dyeing, Tunel method observation after acupuncture treatment of retinal functions and organizational form,ultrastructure and photographic cell apoptosis change; The determination of nitrate reductase enzymatic the content of the retina NO; Using xanthine oxidase method to determine the content of SOD the retina; The highly effective liquid phase color spectrometer detection of glutamic acid content; RT-PCR detection technology p53mRNA retina expression.Results:MfERG test results found that group compared with normal, the total ring N1wave and P1wave dives are extended, reaction density are weakened, the difference was statistically significant (P<0.05),14days still not recover,and model group comparison, acupuncture group N1and P1wave reaction were significantly increased,the difference was statistically significant (P<0.05); HE dyed found that group compared with normal, the outer core layer in the retina thinning, inside and outside section arranging disorderly, no clear boundary, the nucleus swelling, section in mitochondria swelling and the quarter plate membrane rupture, and especially the granular layer and light sensors cell layer almost disappeared, and the model group compared, acupuncture group still retain have2-4layer outer core layer and a certain number of photoreceptor cells; Tunel method observation, compared with normal group,the average density of light are increased,indicating that the photoreceptor cells occur apoptosis,the difference was statistically significant (P<0.05); Group compared with model,acupuncture group average light density was significantly lower, prompting the photographic cell apoptosis reduce, the difference was statistically significant(P<0.05); Each group Glu and NO,compared with normal group,content are rising, the difference was statistically significant(P<0.05),and compared with model group, acupuncture group the Glu and NO content is reduced and the difference was statistically significant (P<0.05); The SOD vitality,compared with normal group,content are lower, the difference was statistically significant (P<0.05), and compared with the model group, acupuncture group SOD vigor significantly higher, the difference was statistically significant (P<0.05); Compared with normal group, each group P53express all rise,the difference was statistically significant(P<0.05), and the model group compared, acupuncture group P53express drop, the difference was statistically significant (P<0.05)。Conclusion:Acupuncture may a certain degree MNU inhibition of apoptosis induced photoreceptor cells, promote the recovery of visual function, reduce the retina tissue cell damage and its mechanism and inhibit rat retina P53mRNA expression, improve the retina SOD activity and reduce glutamic acid and NO content thus inhibiting photographic apoptosis and oxidative stress reaction relevant.
