Salvianoic acid B is one major (about70%) and most efficient component of water-soluble salvianoic acid from Chinese medicinal plant Salvia miltiorrhiza. The related research results showed that it has potent pharmacological effects such as anti liver injury, reduction of atherosclerosis, myocardial protection, anti-tumor, treatment of pepticulcer, and memory dysfunction improvement, prevention and treatment of neurodegenerative diseases. Because its concentration is low and with many other complicated components in the salvia miltiorrhiza, purification by chromatography technique is cumbersome and high in cost, and find its limitations in practical application to get salvianolic acid B from other water-soluble compounds of salvia miltiorrhiza. In recent years the research about salvianoic acid B synthesis has been carried out all over the world, as an important intermediate of salvianoic acid B, the synthesis of dihydrobenzofuranoid compound can provide important basis and theoretical significance.In this paper, with3-Hydroxy-p-anisaldehyde as starting material, to prepare some compounds sharing the skeleton of salvianoic acid B, the synthesis routes have not been reported.In the first route, followed successively by its acetylation, Fries rearrangement and Willgerodt rearrangement to prepare some compounds sharing the skeleton of Salvianoic acid B.The effects were optimized by molar ratio, catalyst, time and temperature on yield. And its structure was characterized by1HNMR and MS.In the second route, an important intermediate of Salvianolic Acid B, dihydro-benzofuranoid was synthesized from3-hydroxy-4-methoxybenzaldehyde by a six-step reactions of bromide, esterification, acetalization, nucleophilic reaction, hydrolysis and cyclization, in total yield of21.4%.Its structure was characterized by HNMR and MS. The effects were inpecteted by catalyst, molar ratio, resolve, time and temperature on yield, and the yield was improved.
