BackgroundLung cancer is one of the most common and deadly cancer in the world. Studiesshow that the invasions and metastases are the sign of biology and malignancy, anddistant metastases are the major causes of death in the patients with lung cancer. A largenumber of studies have found the expression of immunoregulatory protein B7-H3inlung cancer was closely associated with the tumor metastasis (lung cancer), but theexact mechanism is remained to be explored.AimThis study aimed to explore the role of B7-H3in the metastasis of lung cancer,which laid the foundation for the research of the molecular mechanism of B7-H3intumor metastasis.Method(1)The B7-H3shRNA was used to transfect the lung cancer cell H1299, and weconfirmed that silencing of B7-H3, via the fluorescence microscope, real time PCR,RT-PCR and FCM.(2)We applied the test of cck8, scratch, transwell and apoptosis to investigate therole of B7-H3in the proliferation, invasion, migration and apoptosis of H1299, via theB7-H3-H1299cell (H1299-B7-H3-siRNA) and B7-H3+H1299cell (H1299-sn-siRNA).Meanwhile, we explore the role of B7-H3in the progress of lung cancer via in vivoanimal experiments, using the B7-H3-H1299and B7-H3+H1299for subcutaneousvaccination in nude mice.Result(1) The steadily transfected B7-H3konckdown/H1299was constructed, and the mRNAand protein expression of B7-H3in H1299-B7-H3-siRNA was significantly decreased,even could not be detected, compared to the untransfected group. (2) In contrast to B7-H3+H1299, an inhibit on the proliferation, invasion, migrationand increase of apoptosis on B7-H3-H1299were showed.(3) In vivo animal experiments showed that B7-H3knock down tumors displayed aslower growth rate than the control.ConclusionCell B7-H3-H1299was successfully constructed with the silencing of B7-H3inhuman lung cancer cell H1299, the over expression of B7-H3in lung cancer canimprove proliferation, migration, invasion ability and anti-apoptotic ability, in vivoanimal experiments showed that B7-H3promotes the proliferation and the invasion andmetastasis of lung cancer cell.
