| Aralia elata (Aralia elata Seem.A.mandshurica Max), also as known as Liaodong Aralia, commonly known as "the barbed old Shoots",belong to Araliaceae,is a kind of local herbs that can be easily found in Jilin Province, what’s more,the Aralia elata is rich in amino acids^vitamins^minerals, which also have higher dietary property and medicine property. Aralia elata Monomer saponins (CE) were isolated from Aralia elata total saponins combined with macroporous adsorptive resins and chemical conversion as the compounds oleanolic of acid-3-O-beta-D-pyran glucuronide.Our laboratory’s research has already demonstrated that CE plays an important role in treating arrhythmia disease and inhibiting apoptosis.This study is regarded to set up the experimental animal models of arrhythmia and apoptosis. Thus, the mechanism of arrhythmia can be connect to the apoptosis theory,which can also explore the the mechanism of arrhythmia.we can observe the function of CE in the experimental animal models mention above. the research can also be meanful to develop a new drug of anti-arrhythmia and the anti-arrhythmia mechanism of CE according to morphology analysis of apoptosisã€enzymology analysis of serum(CKã€LDHã€AST)ã€the percentage of apoptosisã€the expression of the gene of relating to apoptosis. The study was divided into two parts:1ã€the establishment of animal models related to arrhythmia and apoptosisThe female Wistar rats weighting220-240g were randomly divided into normal control group and model group1, model group2ã€model group3,then, the rats were injected lg/kg ethylurethanm in intraperitoneal to record the normal ECG of all the rats.when the rats awake,the control group of clearing level were infused with10ml/kg·d water and the model group were infused with1.46mg/kg·d aconitine once a day respectively. the model groups were separately given aconitine3daysã€7daysã€10days, The rats were sacrificed after given the last water or the aconitine48hours, and hearts were collected to put in10%formaldehyde solution to detect the percentage of cardiac muscle cell apoptosis by HE. so the method of establishing the animal models related to arrhythmia and apoptosis can be definited。The female Wistar rats weighting220-240g were randomly divided into normal control group and model group, the normal ECG of all the rats should be taken and recorded.then,when the rats awake,the rats of control group were infused with10ml/kg·d water and the rats of model group were infused with1.46mg/kg·d aconitine once a day respectively. continuously10days. The ECG of all the rats should be taken and recorded after given water or the aconitine the first time.The rats were sacrificed after given the last water or the aconitine48hours,It’s necessary to measure the activity of CKã€LDHã€AST of serum after the blood was centrifugated, and the hearts were collected to detect the percentage of apoptosis by TUNEL, and observe the ultra-structure of cardiac muscle cell by TEM.the result was found that the animal models related to arrhythmia and apoptosis was founded by the method of giving the rat aconitine1.46mg/kg for10days.2ã€exploring the intervention of CE on the experimental animal models of arrhythmia and apoptosisThe femal Wistar rats weighting220-240g were randomly divided into normal control group, model group, amiodarone group (55mg/kg),high concentration of CE group(15mg/kg), middle concentration of CE group(7.5mg/kg), low concentration of CE group(3.75mg/kg), then, the rats were given the ethylurethanm, collecting the normal electrocardiogram when the rats in anesthesia state, when the rats wake up,the rats of clearing level except normal group were infused with1.46mg/kg·d aconitine once a day for10days respectively, the rats were continuously given aconitine in the ante meridiem and the medicine in the post meridiem. after giving the rats aconitine the first time, the electrocardiogram of the rats were collected and recorded, the rats were given medicine for12days. after giving the rats the last medicine48hours,the rats were given1.0g/kg ethylurethanm,then, the blood was collected to centrifugalize, the serum was used to detect the activity of CKã€LDHã€AST; besides,the hearts were put in the4%paraformaldehyde solutions after perfusion of abdominal aorta, which were used to detect the percentage of apoptosis by TUNEL method,the hearts were also used to observe the ultra-structure of cardiac muscle by TEM, what’s more, hearts were collected to put in-80℃fridge to detect the expression of protein related to apoptosis. The results indicate: CE plays an important role in inhibiting the apoptosis relating to decrease the CKã€LDHã€AST of serum in injured cardiac muscle cell,the result of TEM:ultra-structure of cardiac muscle cell of model group was tremendous injuried than control group.while amiodarone group and the three dose group of CE can decrease the injury of cardiac muscle cell,the ultra-structure of cardiac muscle cell of high concentration of CE group was mostly closely to control group. more over, the function of protecting the cardiac muscle cell is related to up-regulation the expression of Bcl-2ã€inhibiting the activation of caspase-3ã€caspase-9, uncorrelated with expression of Bax. the protecting the cardiac muscle cell of CE may through the ERK-MAPK channel. |
