ObjectiveTo observe follistatin. activin A and BMP-4expression in different cerebral regions during the brain development of rat.MethodsThirty simultaneous be pregnant Sprague-Dawley rats, were randomly divided into six groups, five rats every group:E8.5、E13、E18、P3(was born three days later)、P7、P30. Five embryos every pregnant rat was fixed and P3、 P7、P30rats cerebral perfusion fixed and section contain the cortex、striatum、 hippocampus and olfactory bulb for Immunohistochemical. Five embryos every pregnant rat and P3、P7、P30rats cerebral of the cortex. striatum. hippocampus and olfactory bulb was also used to abstract follistatin mRNA activin A mRNA and BMP-4mRNA on the ice table for Reverse transcripttion polymerase chain reaction (RT-PCR). Expressions of follistatin、activin A and BMP-4in the cortex、striatum. hippocampus and olfactory bulb were examined by means of ABC immunohistochemical method under light microscope. Reverse transcripttion polymerase chain reaction (RT-PCR) method was also used to investigate the expression level of follistatin mRNA. activin A mRNA and BMP-4mRNA in brain during development.ResultsFollistatin and activin A expression in rat brain was strong in embryonic day8.5, but in embryonic day13, the number of positive cells was decreased and in postnatal month1was distinctly decreased. In the same phase, there are not distinct differences patterns in different tissues. BMP-4expression in rat brain was wide, the positive cells in the observed regions were abundant, in the cortex and striatum were stronger in the same phase, but from embryonic day8.5to postnatal day7appeared with a lower level, especially showed extremely weak in the hippocampus. From the results of RT-PCR, the peak of follistatin mRNA and activin A mRNA expression was found in E8.5rat brain, decreased in E13, decreased gradually from embryonic day8.5to postnatal1W during the developmental stage, the lowest level in postnatal1M, In the same phase, there are not distinct differences patterns in different tissues. The expression level of BMP-4mRNA in rat appeared lower from embryonic day8.5to postnatal day3during the developmental stage, increased in postnatal1W, the peak was revealed in postnatal1M, in the cortex and striatum were stronger in the same phase.ConclusionThere are expressions of follistatin mRNA. activin A mRNA and BMP4mRNA in the cortex、striatum、hippocampus and olfactory bulb in rats at different developmental stages. The expression level is closely correlated with the developmental age. This indicates that follistatin. activin A and BMP4play important roles in the development of rat cortex. striatum. hippocampus and olfactory bulb.
