| ObjectiveTO discuss the hUCMSCs and VEGF combined transplantation in thetreatment of cerebral ischemia in rat, to investigate its promotion of functionrecovery and angiogenesis, to lay a experimental foundation for the clinicaltreatment.MethodsPurificated hUCMSCs were cultured in vitro. The rats were induced to focalcerebral ischemia models with middle cerebral artery occlusion (MCAO). Fivegroups were arranged:1) MCAO group (no treatment);2) sham group, the leftcommon carotid artery were separated and gave no treatment;3) hUCMSCsgroup (hUCMSCs were transplanted focally);4) VEGF group (VEGF wastranlpanted focally;5) combined group (combined transplanted of hUCMSCs andVEGF focally). Preparing the3rd passage hUCMSCs labeled by Brdu with theconcentration of1×106/ml, VEGF with the concentration of1ng/μl beforetransplantation. After MCAO24h three-dimensional controller was equipped localventricle transplant to striatum and cortex ischemia area. After MCAO1ã€4ã€7ã€14ã€21ã€28d, mNSS to detecet the function recovery of the rat; after MCAO28d,the changes of histological were compared by the staining of Nissl; afterMCAO7ã€14ã€28d, volume of infarction was detected by triphenyltetrazoliumchloride (TTC) staining, the expression of BrdU and vWF were detected by immunohistochemistry technologies,and then the MVD calculation.Results1The mNSS scores: except the sham group, there were significantdifference, the combined group has a better function recovery than the othergroups at28d.2The Infarct volume: except the sham group, the infarct volume decreasetime dependently, the rest groups show a significant difference(P<0.05), thecombined group has a least volume of infarction at the28d.3BrdU+cells: the BrdU+cells of the combined group and the hUCMSCsgroup show a siginifcant difference(P<0.01); the combined group has moreBrdU+cells in the lesion area(P<0.01).4The MVD: the MVD in the groups of various treatment factors and timeshave significant differences(P<0.01), the combined group has a better MVD thanthe other groups at28d.5Nissl staining: in the sham group,the morphology of the neural cells werecompleted;while in the MCAO group, the nissl of the neural cell disappeardiffusely, the residue nissl bodies show a unbalance distribution.ConclusionsBoth hUCMSCs and VEGF have the function of promoting angiogenesis,improve recovery of the neurological functions in MCAO rat, synergistic effect canbe produced by the combined transplantation, it is better for the injury recovery ofischemia brain. |
