ObjectiveTo cultivate cardiomyocyte of SD neonate rats in vitro,then observ theeffect of Apelin-13on ischemia-induce cardiomyocyte autophagy in glucosedeprivation(GD) neonate rat.MethodsTo cultivate cardiomyocyte of SD neonate rats in vitro.Then qualificate thepurity of cardiac muscle cell.Set up the models of ischemia-inducecardiomyocyte, then group and experimentize. Add interfere factors for eachgroup, terminate the test after12hours.Observe the quantity of autophagybody and autophagy vesicles by electron microscopy.Estimates the ratios ofautophagy body compare with the cytoplasm.ResultsCompared with the normal control group,there are more autophagy bodysand autophagy bubbles in GD model groups,and the ratios of autophagy bodycompare with the cytoplasm are increscent (0.02060.00598,0.08740.00740,P<0.01).Compared with GD model groups,the number ofautophagy bodys and autophagy bubbles and the ratios of autophagy bodycompare with the cytoplasm are all reduced in the group of high and middledose Apelin-13(0.05600.00387,0.06580.00873,P<0.05).The group oflow dose Apelin-13has no significance effect in the autophagy. ConclusionThe autophagy resides in normal myocardial cells.Glucose deprivationcan aggravate it.The Apelin-13of ectogenesis will inhibition it.The finhibitoryaction is relating with concentration of Apelin-13.The autophagy can beinhibited with the three concentration,ang the action raised with theconcentration heighten.
