ObjectiveTo observe the effects of As2O3(As)on the number and function of skinLangerhans cell (LC) through mouse epidermal single cells treated with As,aiming at the mechanism of As to treat skin disease and the pathogenesis ofLC related skin disease.MethodsEpidermal single cell suspension was prepared from C57BL6mice, treatedwith As of different concentration. Appropriate concentration was investigatedusing MTT method. After co-cultured with As, epidermal single cells werestained with appropriate fluorescence conjugated antibodies, and theexpression of co-stimulators of CD80, CD86was analyzed by using flowcytometry to see the effect on LC maturation. FITC-Dextran uptake experimentwas performed to identify the effect of As on ability of LC phagosytosis.Results1. After treated with As for48h, the total cell number was slightlydecreased from2uM, and decrease more with the increase of the concentration.2. After treated with2uM As, the relative ratio of LC was significantlydecreased from1.11±0.12to0.99±0.13, with a p=0.04293. After treated with2uM As, skin LC was separated into two subgroups ofMHC IIhigh,MHC IIlow. Further gating MHC IIhighpopulation, the meanfluorescence intensity of MHC II was increased, and the Langerin expressionwas decreased. 4. After treated with2uM As, based on CD80expression, LC showedCD80+and CD80-population, however, based on CD86expression, LCshowed three populations of CD86-、 CD86low, and CD86highLCs. Thepercentage of CD80+and CD86highLCs was decreased, but the percentage ofCD86lowLC was significantly increased. Simultaneously, the MFI of CD80wasdecreased, with the MFI of CD86in CD86lowLCs remained the same.5. After treated with2uM As, MHC IIlowLC population lost function ofantigen phagocytosis, and the ability was impaired in MHC IIhighLCs too.Conclusion1. As affected the percentage of LC in skin cells.2.2uM As affected skin immunity through affecting the expression of MHCII and Langrin.3.2uM As affected skin immunity through affecting LC maturation ability.4.2uM As affected skin immunity through affecting LC antigenphagocytosis.
