Experiment Objective:Found the model of the Hepal-6liver cancer on C57BL/6rats, and observe the effect of sport on the treatment of the liver cancer via tumor targeting drug delivery system, to explore the mechanism of sport affection on tumor targeting drug delivery system’s liver cancer treatment.Experiment Methods:Tumor drug targeting vector material mPEG-PLGA-PLL-cRGD was combined in experiment1. Drug encapsulation efficiency (EE) and loading content (LC) of the nanoparticles wrew evaluated by ultraviolet spectrophotometer. In experiment2,78male C57BL/6rats with Hepal-6liver cancer are divided randomly into six groups:FREE、SP、 DHAQ、NPS、SP+DHAQ、SP+NPS. Rats were intraperitoneally administered with DHAQ or DHAQ-loaded nanoparticles, then exercise groups performed treadmill running for14days. The weight of the rats and the tumor volumes were measured every two days. At the end of the exercise, the amount of NO、H2S were measured in group FREE、SP、SP+DHAQ、SP+H2S.Experiment Result:1. The tumor drug targeting vector material mPEG-PLGA-PLL-cRGD was combined successfully.2. Exercise was limited in the treatment of liver cancer. In addition as an adjuvant treatment, exercise increase the curative effect of DHAQ-loaded nanoparticles.3. After14days exercise, the mount of serum NO in group FREE was obviously lower than group SP、SP+DHAQ、SP+NPS.4. After14days exercise, the mount of aorta H2S in group FREE was obviously lower than group SP、SP+DHAQ、SP+NPS.Conclusion:1. Proper exercise can strengthen the vector material mPEG-PLGA-PLL-cRGD’s target distribution for liver cancer.2. After proper exercise, the amount of NO and H2S in rats of group SP、SP+DHAQ、SP+NPS were obviously higher than that in group FREE. The treatment of group SP and SP+DHAQ was not not better than group FREE. One of the effectual mechanism that exercise reduce the tumor growth speed maybe that exercise can enhance the expression of NO and H2S, what strength the target distribution of vector material mPEG-PLGA-PLL-cRGD.
