Ganoderma duropora Lloyd is dry fruiting body of Ganoderma duropora Lloyd from polyporaceae fungi.It is distributed in China’s Fujian, Guangdong, Hainan. Guangxi, Guizhou and other regions.It is rich in our province.So Ganoderma duropora Lloyd is widely circulated and used in the civil market and medicinal fungi as Ganoderma.The best reflux extraction method is established by the orthogonal design method for physiologically active substances from Ganoderma duropora Lloyd,polysaccharides and total triterpene acid. Firstly,extraction solvent ethanol concentration and alkaline extraction reagent are studied,when the triterpene acid is extracted.And95%ethanol and4%potassium hydroxide is the optimal concentration and alkali extraction reagents respectively.According to orthogonal test,four factors and three levels of each factor,the best reflow extraction conditions for Ganoderma duropora Lloyd triterpene acid are as follows:60℃,3times,25-fold water,2hour.Besides,for polysaccharide from Ganoderma duropora Lloyd,the best reflow extraction conditions are70℃,3times,40-fold water,1hour.Under the optimum extraction conditions,we measured the content of triterpenoid and polysaccharide of different harvest period from Ganoderma duropora Lloyd.The content of triterpenoid acid from different harvested Ganoderma duropora Lloyd are1.98±0.01,2.10±0.04,0.70±0.01mg/g.In addition,the content of polysaccharide are1.25±0.03,1.57±0.02,1.25±0.01mg/g separately.According to the results above,the polysaccharide and triterpene acid from late season of Ganoderma duropora Lloyd is the highest.And it provides the harvest selection for Ganoderma duropora Lloyd.Under the same conditions,the similarity between chromatogram of triterpene acid from different harvested time and the control map generated from chromatogram TCM fingerprint is0.896,0.9,0.834.And according to compare chromatogram of six monosaccharide mixed standard with polysaccharides from different harvested,the monosaccharide composition of early ones contend mannose, glucose, galactose, fucose;late ones contend mannose, ribose,glucose,galactose,xylose and fucose;the third ones contend mannose,ribose, glucose, galactose, and fucose.In summary,different harvest time has great effect on the content and composition of triterpene acids and polysaccharides from Ganoderma duropora Lloyd.Polysaccharides from Ganoderma lucidum has the effect of antioxidation.To determine the antioxidant activity of polysaccharide from different harvested of Ganoderma duropora Lloyd,we studied four antioxidant models in vitro,containning superoxide anion scavenging activity,DPPH free radical scavenging,ABTS free radical scavenging,reducing power.Polysaccharides from different harvested of Ganoderma duropora Lloyd have different antioxidant activity.The ic50of DPPH free radical scavenging are99.05,86.83,101.06ug/mL separately. ABTS are1.39,0.93,1.57mg/mL.Superoxide anion scavenging are0.52,0.27,0.38mg/mL respectively.Through statistical analysis,polysaccharide from Ganoderma duropora Lloyd has effect on antioxidation.Besides,late season polysaccharides from Ganoderma duropora Lloyd is significantly higher than that in early season and the third season.But there is no significant difference in the antioxidant activity of early season and the third season.In consequence,different harvest time has an effect on the antioxidant activity.We have separated nine compounds.And seven are firstly isolated from Ganoderma duropora Lloyd.There are certain difference on the amino acid composition and ion exchange chromatography of the different harvested fruiting bodies from Ganoderma duropora Lloyd.This can be used as one of quality evaluation index.In conclusion,there are few studies on Ganoderma duropora Lloyd.And its chemical composition and biological activity are worthy of being researched in depth.
