Chronic cerebral ischemia is a group of diseases which is common in the nervoussystem. It is also the common pathological basis of Alzheimer’s disease、 vasculardementia and other diseases. Cognitive impairment is the main clinical situation in theearlier period of chronic cerebral ischemia. and then it cause the permanent neuralimpairment, of which the pathological mechanism is different from acute cerebralischemia. Many of the mechanisms of injury, such as energy metabolism, immuneinflammatory injury, apoptosis, oxidative stress, etc maybe play a role in chroniccerebral ischemia. Which is mostly concentrated on Oxidative Stress in acute cerebralischemia or ischemia-reperfusion, little study of the role of oxidative stress in chroniccerebral ischemia. Brain tissue is sensitive to the damage of oxygen radical. Therefore,the study of the correlation between the chronic cerebral ischemia and oxidative stressis instructive to exploring the relationship of chronic cerebral ischemic injurymechanisms, and other related diseases, Which is also helpful of the using ofclearing oxygen radicals drugs and other exogenous antioxidants.Objective:1、Observed the varying levels of NO and NOS in chronic ischemia rat braintissue.2、 provided guidance on prevention and treatment of chronic cerebralischemia from the perspective of oxidative stress clinically.Methods:1、Thirty-six Wistar male rats were divided into three groups randomly:1daygroup、7days group、30days group. Each group was divided into sham group,ischemia group.2、Produced a model of chronic cerebral ischemia through the method ofpermanent bilateral carotid artery ligation3、Determined the content of NO in the brain tissue of rats in each group at each time point with Enzyme-linked immunoadsordent assay (ELISA).4、Determined the content of NOS in the brain tissue of rats in each group at eachtime point with Western blotting (Western blot) at the protein level.Result:1、The content of NO and NOS in every experimental rats were significantlyincreased compared to sham-operated rats.2、The content of NO and NOS in the brain tissue of rats gradually increasedfrom1d to7d and then30d.Conclusion:1、the content of NO and NOS in brain tissue have consistency in the processof chronic cerebral ischemia.2、NO and NOS play a role not only in the acute phase of chronic cerebralischemia, but also in chronic phase chronic cerebral ischemia.
