Purpose: This experimental rat model to stress urinary incontinence (SUI)in different parts of injected transplantation of human umbilical cordmesenchymal stem cells (hUCMSCs), research the hUCMSCs role in thetreatment of SUI. Methods: analog birth trauma and post-menopausalbuild a rat model of SUI, the hUCMSCs were transplanted to the proximalsciatic nerve model around the urethra and urethral sphincter, a regularfeeding, by the the urodynamic and analog sneeze experimentaldetectionmaximum bladder capacity (MCC), leak point pressure (LPP),and "analog sneeze" experimental observation of any leakage of urinegenerated to evaluate the bladder urine storage capacity, and then studythe different parts of the hUCMSCs transplant have a therapeutic effect onSUI optimization bestmigration path. Results: The model rats the MCC(maximum bladder capacity), LPP (LPP) are lower than normal group, bycompletely randomized design t-test, p <0.05, statisticallysignificant;When the bladder instillation half of the maximum bladdercapacity, model group increased abdominal pressure87.5%can beinduced leakage of urine, the normal group, only12.5%can induce leakage of urine, by the chi-square test, P <0.05.Within the urethrasphincter group,the experimental group,the experimental group and thecontrol group after local injection of MCC increased in the experimentalgroup increased more,by completely randomized design t-test,pvalue<0.01,the difference was statistically significant;this group,theexperimental group therapy after the LPP increased in the control groupafter injection LPP reduced by completely randomized design t-test,pvalue <0.01,the difference was statistically signifucant.Periurethralgroup,the experimental group and the control group were increased afterlocal injection of MCC,the completely randomized designt-test,p>0.05,the difference was not statistically significant;this group,theexperimental group after treatment LPP increased control group wasinjected after the LPP decreased by completely randomized design t-test,pvalue <0.01,the difference was statistically significant.Sciatic nervegroup,the experimental group and the control group after local injection ofMCC,LPP were increased by completely randomized designt-test,p>0.05,the difference was not statistically significant.Tumorigenicityexperimental observation3.5months without tumorformation.Conclusion:hUCMSCs within the urethra sphincter injectiontransplant can improve SUI rat urine storage capacity,short-term effect issignificant;would hUCMSCs injected into the within the urethra sphinctertransplant can improve faster SUI rat urine storage capacity;periurethral transplantation can improve SUI hUCMSCs urethral closure pressure inrats,but can not improve SUI rat urine storage capacity;sciatic nervetransplant hUCMSCs short term injection of rat urine storage capacity SUIno improvement.
