Objective:Observing changes the in the behavior of chronicmethamphetamine (MAP) dependence rats, which is the resulted fromthe intervention of z-DEVD-fmk. Observing activity changes ofCaspase3in brain cells and apoptosis of relating tissue of rat brain aswell as its influence on expression of BCL2protein.Method: Establish chronic methamphetamine (MAP) dependencerats as experimental animal model. Then implement the intervention ofinjecting z-DEVD-fmk into the lateral ventricle of rats. Using enzymestandard meter method to detect Caspase-3activity.Using TUNELmethod to detect the apoptosis of relating tissue of rat brain andimmunohistochemical technology to test the effects of the BCL-2protein expression.Results:1. The percent of MAP chronic dependent rats which showsabnormal behavior was100%. The mainly abnormal behavior includinginvoluntary nod, piloerection、self-mutilation、circles、irritability andsome other symptoms. These symptoms occur less after givenz-DEVD-fmk.2.Activity of Caspase-3enzyme is getting lower after givenz-DEVD-fmk compared with MAP group (P<0.05).3.The apoptosis number of relating tissue of rat brain wassignificantly decreased after given z-DEVD-fmk compared with that inMAP group(P<0.05). 4.The number of BCL-2positive cells was increased significantlyafter given z-DEVD-fmk compared with MAP group (P <0.01).Conclusion:1.The activity of Caspase-3is getting lower after the inhibition ofz-DEVD-fmk compared with MAP chronic dependence rats.2. The number of apoptosis of relating tissue of rat brain of the MAPchronic dependence rats is getting lower after the inhibition ofz-DEVD-fmk and expression of BCL-2is increasing at the same time.
