Objective:Explore the expression of Eag1potassium channel、hypoxiainducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF)in different cervical lesion tissues, tumour grade, clinical staging and lymphnode metastasis and radiotherapy of cervical squamous cell carcinoma. Studythe correlation between Eag1、HIF-1α and VEGF. Looking for new predictivemarkers for patients with cervical squamous cell carcinoma for diseasediagnosis and prognosis. Observe the effects on Hela cells of linearaccelerator radiation after the use of potassium channel blocker4-AP. Providea theoretical basis in improving radiation effect in the patients with cervicalsquamous cell carcinoma.Method:1. Eag1expression、VEGF expression and HIF-1α expressionin different cervical tissues were measured by immunohistochemistry.2. Detect the inhibition rate of hela cell proliferation, apoptosis rate withdifferent concentrations4-AP and different radiation dose by MTT and flowcytometry.Results:1. Immunohistochemistry showed that the expression ofEag1、HIF-1α and VEGF protein in cervical squamous cell carcinoma tissueswere significantly higher than the chronic cervicitis group and the cervicalintraepithelial neoplasia group(P<0.01). The expression of Eag1、HIF-1αand VEGF in non-keratinizing squamous cell carcinoma were higher than thekeratinizing squamous cell carcinoma group (P<0.05). The expression ofEag1、HIF-1α and VEGF in the stage IIb-IV group were higher than IIb and above period group (P<0.05). These three proteins expression in the lymphnode metastasis of cervical squamous cell carcinoma were higher than thosewithout lymph node metastasis group (P<0.05). These three proteinsexpression before radiotherapy in cervical squamous cell carcinoma washigher than post-radiotherapy group (P<0.05). The expression of Eag1、HIF-1α and VEGF in cervical squamous cell carcinoma was positivelycorrelated.2. Rate of Hela cell proliferation and apoptosis rate increased along withincreasing of radiation dose and the concentration of the potassium channelblocking agents(4-AP). Proliferation inhibition rate and apoptosis rate ofUnited Group (radiation+4-AP) was higher than alone4-AP group andradiotherapy group. Hela cell proliferation inhibition rate and apoptosis ratewas positively related to potassium channel blockers (4-AP) concentrationand the radiation dose.Conclusion:1. The Expression of Eag1、HIF-1、VEGF were associatedwith cervical carcinoma degree lesion. The express of this three proteinincreased as the development of clinical stage and the decreased of pathologicgrade in cervical cancer. The express of this three protein increased afterlymph node metastasis.The express of this three protein decreased for thepaitient after radiation therapy.The expression of Eag1、HIF-1α and VEGF incervical squamous cell carcinoma was positively correlated.Eag1、HIF-1αand VEGF protein play an important role in the development of cervicalcarcinoma. Eag1、HIF-1α and VEGF can be used to predict prognosis incervical squamous cell carcinoma.2. Potassium channel blocker4-aminopyridine (4-AP)can significantlyincrease the radiation of Hela cell proliferation inhibition rate and apoptosis rate, enhance the radiotherapy effect. This will provide new ideas for theresearch of advanced cervical carcinoma radiotherapy.
