| Lo Han Kuo, the dried fruit of the plantSiraitiagrosvenorii(Swingle) C·Jeffrey exA·M·Lu etZ·Y·Zhangwhich belongs to Cucurbitaceae family Siraitia genus, is used asboth drug and food. It is recorded in Chinese Pharmacopoeia PartOne. Lo Han Kuo has a function of Clearing heat and moistening lung,relieving sore-throat to restore voice and relaxing bowel[1]. Modernpharmacological analysis indicates that extract of Lo Han Kuo hasbiological activity as hypoglycemia and anti-oxidation[2][3]. Forfully making use of the source of Lo Han Kuo, Our laboratory combinedHubei Hui Te Biomedical Technology Ltd, co. to realize thecombination of industry and research. We systemically study onpharmacodynamic substance foundation of stem and leaf in Lo Han Kuo.The active component part is flavonoid, it has an efficienttherapeutic action in antibiosis, anti-oxidation, and non-insulindependent diabetes rats model. The main active component isKaempferol3,7-L-dirhamnoside.In further explanation of pharmacodynamic material basis oinLo Han Kuo stem and leaf, we studied in serum medicinal chemistryof the extract from Lo Han Kuo’s active part. We determined theblood pharmaceutic components of Lo Han Kuo stem and leaf extracts by comparing the HPLC fingerprints of Lo Han Kuo stem and leafextracts, drug serum and blank serum. Result indicated15peaks weredetected in Lo Han Kuo stem and leaf extracts by HPLC detection,seven of them were drug original components and one was Kaempferol3,7-L-dirhamnoside. This study provides an favorable reference fordetermination of flavonoid as the active component in Lo Han Kuo’s stem and leaf.We used SD rats to study pharmacokinetics of Kaempferol3,7-L-dirhamnoside to analyze the metabolic pattern of the activecomponent in Lo Han Kuo’s stem and leaf. Single extravascularadministration was used to collect orbital blood of rats at regulartime. The concentration of blood drug in different time wasstatistically used for standard curve preparation. DAS software wasused to analyze the fitting data and to calculate thepharmacokinetics parameters. The results indicated the absorptionand elimination of Kaempferol3,7-L-dirhamnoside in male rats arefaster. The concentration-time curve was a bimodal curve. Thispaper discuss the metabolic pattern of Kaempferol3,7-L-dirhamnoside, which is the presentative component of extractin Lo Han Kuo, and provide reference for development and applicationof stem and leaf in Lo Han Kuo as a Chinese medicine product. |
