Objective: To investigate conventional ultrasound characteristics of VX2tumorsin fatty1iver and normal liver of rabbits. Analysis the VX2tumors in fatty1iver andnormal liver of rabbits blood perfusion characteristics with contrast-enhancedultrasound (CEUS), which quantitative parameters correlation with the tumorangiogenesis.Methods: Rabbit models of fatty1iver and normal liver with implanted VX2tumors were established, furthermore, examined using conventional and CEUS. Toinvestigate the conventional ultrasound characteristics and CEUS blood perfusioncharacteristics of VX2tumors. Two groups of hepatic backgrounds and VX2tumorswere analyzed by QontraXt quantitative software of CEUS. The expression level oftumor MVD and VEGF were detected with Immunohistochemical techniques. Theexpression level of tumor VEGF gene were detected with realtime fluores-cencequantitative PCR (FQ-PCR).Resluts: The fatty liver models with20VX2tumors and normal liver modelswith23VX2tumors were successfully developed in rabbits.(1)VX2tumors in fatty liver, iso-echo and hypo-echo were seperately9/20(45%),11/20(55%) in conventional ultrasonography. There was no significantdifference of VX2tumors in normal liver in conventional ultrasonography. Comparedwith the normal liver tumors, incidence rate of hyperechoic lesions in fatty liver VX2tumors were lower in conventional ultrasonography (P<0.05).(2)CEUS shows that the VX2tumors were typical pattern “fast-in and fast-out”.CEUS showed arterial phase enhancement in fatty liver VX2tumors,5-12s in arterialearly phase rapid increase to peak (7of them for hemogeneous enhancement;13ofthem for the ring enhancement). Then about14-27s tumor begin to hypoenhancement quickly. In normal liver VX2tumors,6-11s arterial early phase rapid increase to peak(12of them for hemogeneous enhancement,11of them for the ring enhancement).Then about16-24s tumor begin to hypoenhancement quickly.(3)①Fatty liver background compared with normal liver background, arrivaltime of contrast agent was late, peak intensity was low (P<0.05). The expression ofVEGF gene in fatty liver parenchyma are lower than normal liver parenchyma(P<0.05).②Fatty1iver VX2tumors compared with normal liver VX2tumors.Arrival time of contrast agent, peak intensity, sharpness, area under curve had nosignificant difference between the two groups. The MVD and expression of VEGFhad no significant difference between VX2tumors in normal1iver and fatty1iver(P>0.05). No significance was found in VEGF gene expression in VX2tumors innormal1iver and fatty1iver.(4)①The arrival time of contrast agent, time to peak in fatty liver VX2tumorswere early than the surrounding liver parenchyma and peak intensity, area undercurve compared with peripheral parenchymal were significant different (P<0.05).The sharpness had no significant difference between the two groups(P>0.05).②Thearrive time, time to peak in normal liver VX2tumors were early than the surroundingliver parenchyma, and peak intensity, sharpness, area under curve compared withperipheral parenchymal were significant different (P<0.05).(5)There was positive correlation between MVD and peak intensity (r=0.494,P<0.05) in fatty1iver VX2tumors, but MVD showed no correlation with arrival time,time to peak, area under curve. The expression of VEGF were not correlated with TICparameters (all P>0.05).MVD had positive correlation with expression of VEGF(r=0.508,P<0.05).There was positive correlation between MVD and peak intensity (r=0.591,P<0.01) in normal1iver VX2tumors, but MVD showed no correlation with arrivaltime, time to peak, sharpness and area under curve. The expression of VEGF were notcorrelated with TIC parameters (all P>0.05). MVD had positive correlation withexpression of VEGF (r=0.57,P<0.01). Conclusions:(1)There is significant difference in characteristics of conventionalultrasonography between fatty1iver and normal liver VX2tumors. Compared withthe normal liver, incidence rate of hyperechoic lesions in fatty liver VX2tumors werelower in conventional ultrasonography.(2)CEUS blood perfusion in fatty liver was lower than that in normal liver. Theexpression of VEGF gene in fatty livers were lower than those in normal livers.(3)CEUS quantitative parameters, the expression of MVD and VEGF had nosignificant difference between fatty1iver and normal liver VX2tumors.(4)The quantitative parameters of CEUS had positive correlation with MVD infatty1iver and normal liver VX2tumors, which can indirectly reflect the MVD andmay be helpful to evaluate tumor angiogenesis....
