Background:To investigate the expression of PAUF and β-catenin in colorectalcarcinoma and the effect of PAUF RNA interference on cell proliferation, apoptosis, cellcycle progression, invasion,adhesion and migration in the HCT116cell line.Methods:PAUF and β-catenin expression levels were examined by real-time PCR andimmunohistochemistry in colon cancer tissues and adjacent normal tissues.The HCT116cell line was transfected with small interfering RNA (siRNA) targeting on PAUF.Successful transfection was confirmed using RT-PCR and Western blot. Cell proliferationwas measured by MTT assay, and the apoptosis and cell cycle distribution were analyzedby flow cytometry analysis (FACS).Changes in invasion,adhesion and migration of theselected cell line were evaluated by Transwell test,Cell-adhesion assay and Cell-migrationassay respectively.Results:The expression levels of PAUF and β-catenin in adjacent normal tissues werelower than in tumor tissues (P <0.05). Moreover, the expression of PAUF was correlatedwith the expression of β-catenin in both tumor and adjacent normal tissues (P <0.05).PAUF-siRNA transfection down-regulated the expression of PAUF in colon cancer cells,which was also associated with a decrease in the expression of β-catenin (P <0.05).PAUF-siRNA inhibited the proliferation of colon cancer cells, promoted theirapoptosis and arrested these cells in the G2/M phase, and at the same time,PAUF-siRNAinhibited the invasion, adhesion and migration of the tumor cell (P <0.05).Conclusions: PAUF and β-catenin were expressed in colon cancer at a high frequency.PAUF specific siRNA transfection down-regulated the expression of PAUF and β-cateninin colon cancer cells, Moreover, PAUF specific siRNA inhibited the proliferation of coloncancer cells, promoted their apoptosis and arrested these cells in the G2/M phase, and atthe same time, PAUF specific siRNA inhibited the invasion, adhesion and migration of the tumor cell.Interference of PAUF might be an effective strategy through β-cateninregulation of colon cancer.
