Expression Of Thymosin Beta4on Renal Tissues With Mesangial Proliferation And Tubulointerstitial Fibrosis

Objective To investigate the expression and significance of Tβ4on children renaltissue with mesangial proliferation, on folic acid induced nephropathy (FAN) withtubulointerstitial fibrosis. The role and the possible underlying mechanism are alsoresearched in this study.Methods (1)36children confirmed by renal biopsy with main historical changes ofmesangial proliferation were selected from Children’s Hospital Affiliated to SoochowUniversity in the past half a year. They were divided into three groups according to thedegree of mesangial proliferation and two groups based on whether or not they hadtubulointerstitial fibrosis. Immunohistochemistry stain (IHC) was used to detect theexpression of Tβ4. Based on the clinical data of mAlb，UCr，α-MG,β-MG and24hUTP，correlations were discovered among proteinuria, renal pathological changes andTβ4.(2）CD1mouse were randomly assigned to two groups: normal control group andFAN group. The model of interstitial fibrosis was induced with two times of240mg/kgfolic acid (FA) by intraperitoneal injection. Four mice in each group were sacrificed on the14thand21thday. Samples of kidneys were processed for hematoxylineosin stain (HE),Masson trichrome stain. Tβ4was semi-quantified by immunohistochemical stain(IHC).All data were analyzed with SPSS17.0by T test, F test and Rank Sum Test.Results （1） The expression of T β4between mild&moderate and severemesangial proliferation had significant difference(P<0.05). The expression of Tβ4washigher in tubulointerstitial fibrosis group than non-tubulointerstitial fibrosis group(P<0.05).The level of mAlb，α-MG,β-MG and24hUTP among groups of mild, moderate andsevere mesangial proliferation had significant difference(P<0.05).（2） Compared with the control group, the expression of Tβ4in FAN group was increased at day14and day21(P<0.05), while no significant difference between their time point in FAN group(P>0.05).Conclusion (1) The expression of T β4was upregulated in severe mesangialproliferation. Tβ4expression was increased with appearance of tubulointerstitial fibrosis.All these indicate that its expression may be responsed to mesangial proliferation andtubulointerstitial fibrosis.(2) The expression of Tβ4on renal tissue of tubulointerstitialfibrosis was more dominant than control group in FAN mice. Tβ4might play in theprocess of tubulointerstitial fibrosis.(3) The level of mAlb，α-MG,β-MG and24hUTPmay be indicator for the evaluation of mesangial proliferative.
