Objective:To investigate the effects of mesenchymal stem cells from mouse bonemarrow on phenotype of spleen derived dendritic cells and the mechanism of action.Methods:C57BL/6mice bone marrows cells(BMSCs) were isolated in vitro and thenco-clutured with spleen derived dendritic cells of C57BL/6murine at the same ratio forfive days.According to the processing methods, the experiment is divided into two groups,namely control group (A) and experimental group (B).Group A:dendritic cells+lipopolysaccharide (LPS=1ug/ml); Group B:dendritic cells+BMSCs+LPS (1ug/ml),BMSCs:DC=1:1. The morphological of DC were observed by scanning electronmicroscopy and the phenotypes of DC in the two groups were examined by flowcytometry.Results:1. Compared with A group,the expression of ILT4and CD45RB of DCincreased obviously in B group(P<0.05);The expression of CD11c,MHC Ⅱand CD86decreased distinctly(P<0.05);The expression of PDL-1,CD40,CD45RA,CD1d and CD14were no significant difference(P>0.05).2. The number of synaptic of DC in A group ismore than the DC which were untreated with LPS, however, The number of synaptic ofDC in B group is less than the DC which were untreated with LPS.Conclusion: BMSCs can enhance the expression of ILT4and CD45RB and abate theexpression of CD11c, MHC2and CD86; BMSCs can reduce the synaptic of DC and the toinhibit antigen presenting capacities of DC so as to induce the immunological tolerance.
