Objective:To study the protection effect and mechanism of YiQiHuoXue therapy in radiation myocardial injury in mice.Methods:We established an X-radiation injury animal model in irradiating the whole body of the mice with60Coγ ray using the Buyang Huanwu Decoction as supplement intervention control research. And the results were observed by the use of flow cytometry, transmission electron microscopy(TEM), immunohistochemistry(IH) method, RT-PCR method, Western Blot and other methods, which could study prevention of myocardial radiation injury protection mechanism from the organization, protein and molecular level.Results:The results showed that Buyang Huanwu Decoction could improve the myocardial endurance in mice after radiation injury. It has been observed that after received Buyang Huanwu Decoction for14days, the levels of CD3+and CD4+in peripheral blood lymphocytes subsets of the mice in the model group compared with the model group had increased significantly, and the IL-2level in the mice’s serum and spleen had also increased, but each dose of Buyang Huanwu Decoction group was not obvious on the role of CD8+. HE staining results showed that ultimately Buyang Huanwu Decoction could reduce the severity of myocardial radiation damage in mice, especially in the high-dose group significantly. TEM observation showed that Buyang Huanwu Decoction had protective effect on the body. IH staining results showed that the each group of Buyang Huanwu Decoction reduced the severity in mice after irradiation significantly, particularly in the high-dose group. Buyang Huanwu Decoction each dose group of myocardial tissue of mice ATM/ATR-mRNA and Click1/Chek2-mRNA was expressed compared with the model group, there was a certain degree of decline, and the high-dose group decreased more significantly. Buyang five each dose group of myocardial tissue of mice ATM, ATR. Chekl and Chek2protein expression compared with the model group were decreased to some extent, which the high-dose group decreased more significantly, consistent with the RT-PCR method. Conclusions:Radiation damage in mouse myocardium with Buyang Huanwu Decoction has a significant protective effect, especially the high-dose group of Buyang Huanwu Decoction is more obvious. With the deepening of the study of yiqihuoxue method and exploreing the mechanism of pharmacology of Chinese medicine theory, Buyang Huanwu Decoction will get a wider range of applications.
