Objective1. Study the topical haemostasis of the Powder of Total Iridoid Glycosides, the new preparation, from Lamiophlomis Rotata.2. Study the Hydrating Multiplying Factor, Hydrating Velocity and the Tendency of Adhesiveness of the new preparation.3. Study the histocompatibility of the new preparation in topical skin.4. Study the antinociceptive and anti-inflammatory activities of iridoid glycosides from Lamiophlomis Rotata.Methods1. The models of abdominal aorta incision and ears artery cutting in rabbits and the superficial cut of liver in SD adult rats were used to value the topical haemostasis of the Powder of Total Iridoid Glycosides, the new preparation, from Lamiophlomis Rotat. The topical hematostatic effect of the podwer of Aqueous Extract from Lamiophlomis Rotata., Yunnan Baiyao and medicinal starch were compared at the same time.2. The Hydrating Multiplying Factors of the Powder of Total Iridoid Glycosides from Lamiophlomis Rotata, Aqueous Extract from Lamiophlomis Rotata., Yunnan Baiyao and medicinal starch were parallelly determined with the determinator made in our lab. The Hydrating Velocities were parallelly determined in a medicinal stability test chamber with a temperature at37℃and humidity at95%. The Tendency of Adhesiveness of the powders was parallelly determined after absorbing different rates of liquid with the determinator made in our lab.3. Two incisions of back skin were made on each side away from the spine with aseptic operation. One was embedded0.5g powder of Total Iridoid Glycosides or Yunnan Baiyao and another was used as blank control. In the following seven days, the inflammatory reaction of the tissue surrounding the embedment was observed. On the seventh day, the rats were sacrificed and the unabsorbed embedment or the recoved tissue was clip. After the characters of size and hardness being recorded, the tissue was used to pathology test. The infiltration of inflammatory cells and the thickness of fibrous capsule were observed under light microscope4. The hot plate test, the acetic acid-induced writhing test, and the xylene-induced ear swelling test in mice were used to investigate the antinociceptive and anti-inflammatory activities of several iridoid glycosides (200mg/kg i.p.) from Lamiophlomis Rotata.Results1. The average hematostatic times in the model of ears artery cutting were153.75±60.93s,109.88±42.70s,187.50±63.64s, and375.00±71.71s for the powder of the Aqueous Extract, Total Iridoid Glycosides from Lamiophlomis Rotat., Yunnan Baiyao and medicinal starch respectively. The new preparation showed significant topical haematotasis effect (P<0.05). In the model of abdominal aorta incision, the average hematostatic times were5.75±3.82min,3.19±1.62min,8.88±4.59min, and20.19±6.19min in the groups of the powder of the Aqueous Extract, Total Iridoid Glycosides from Lamiophlomis Rotat., Yunnan Baiyao and medicinal starch respectively. The new preparation showed significant topical haemostasis effect (P<0.05). In the model of the superficial cut of liver in SD adult rats, the average hematostatic times were153.75±40.69s,112.50±26.59s,125.2±59.69s,210.00±35.86s for corresponding groups. The new preparation showed significant topical haemostasis effect (P<0.05).2. The Hydrating Multiplying Factors is1.6,1.2and1.2for the powder of Aqueous Extract, Total Iridoid Glycosides from Lamiophlomis Rotata and Yunnan Baiyao respectively, and the medicnal starch can absorb6.2folds water of self-weight. The Aqueous Extract and Total Iridoid Glycosides from Lamiophlomis Rotata showed a rapid Hydrating Velocity than Yunnan Baiyao and medicnal starch. They can get maximum absorption in a very short time. At the same time, the biggest adhesiveness of the powder Aqueous Extract, Total Iridoid Glycosides from Lamiophlomis Rotata and Yunnan Baiyao was higher than medcinial starch and the Total Iridoid Glycosides can get the biggest adhesiveness very quickly.3. After one week, most of the powder of Total Iridoid Glycosides was absorbed and only few infiltration of inflammatory cell could be observed in several mice. But most embedment of Yunnan Baiyao couldn’t be absorbed and formed envelop, even vomica. The infiltration of inflammatory cells and the thickness of fibrous capsule in the group of the powder of Total Iridoid Glycosides were less than Yunnan Baiyao4. Compared to the vehicle group, the iridoid glycosides from Lamiophlomis Rotata could inhibite the response of mice in the acetic acid-induced writhing test, and the edema in the xylene-induced ear swelling test (P<0.05), especially for8-O-acetylshanzhiside methyl ester, 8-deoxyshanzhiside, and7,8-dehydropenstemoside (P<0.01). On the other hand, there was no effect exhibited in the hot plat test (P>0.05).Conclusions:1. The new preparation, the Powder of Total Iridoid Glycosides, has significant topical haemostatic effects in animal experiment modles. This effect is better than the Aqueous Extract from Lamiophlomis Rotata and Yunnan Baiyao.2. The Total Iridoid Glycosides from Lamiophlomis Rotata can absorb liquid and get a strong adhesiveness very quickly. This character is benefit to the topical haemostasis in emergency treatment3. The powder of Total Iridoid Glycosides can be easily absorbed and the histocompatibility of the new preparation in topical skin is better than Yunnan Baiyao.4. Some of iridoid glycosides from Lamiophlomis Rotata showed a moderate antinociceptive and anti-inflammatory activity, which will benefit the treatment of wound.
