| "Wine culture" is an important part of Traditional Chinese Culture of Food and Drink. In China wine is a popular drink, but excessive drinking or long-term alcohol consumption will do adverse effects on people’s stomach, liver, kidney et.al and easily lead to the occurrence of diseases. Meanwhile, it is reported that wine is a serious threat to people’s family happiness and social harmony. This study aims to develop a new kind of effectively Sober-up products according to the market demanded, which was combined Traditional Chinese Medicine extracts with the glucurolactone (glucuronolactone) and to study its mechanism.1. Traditional Chinese Medicine extracts selecting. This study screened the best sober activity of Pueraria lobata40%ethanol extract (P1-e40), and Dendrobium nobile water extract (Dn-w).Screening database of CNKI and Wanfang Data, Pueraria lobata, Wolfiporia cocos, Dendrobium nobile, Radix Glycyrrhizae, Lycium chinense were selected for their better sober activity. The active substances of these medicines were extracted with pure water,40%and80%ethanol respectively, freeze-dried for next experiment. Utilize the traditional rat sober test, the sober activity of each substance were measured. Pl-e40and Dn-w were chose as the best experimental material which could speed upthe sober time of about48.5%and36.1%.2. Sober-up prescription(S-u). It was made up a prescription optimizing the units of Pl-e40, Dn-w and glucurolactone by the response surface methodology in this step. The S-u was constitute with13.67ml/mg Pl-e40,11.99mg/ml Dn-w and0.17mg/ml glucurolactone. With the prescription(S-u), the rats sober up in149.3min in the traditional rat sober test, and the time was shorter about10.4%than the control of "|Jiujialv" which was produced in Yibin Pharmaceutical Co., Ltd. of Wuliangye Group.3. Mechanism study. After gavage with "Erguotou" and S-u, the rats blood were collected3times every hour (twice eyeball blood, last abdominal aortic blood). At the end, the rats were killed and the livers of them were gathered. The serum of blood and liver homogenate were produced, and then the superoxide dismutase (SOD), Alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH), glutathione (GSH) and malondialdehyde (MDA) were detected by kits. The alcohol concentration was also detected by gas chromatograph (GC). Through data analysis, the S-u could improve the blood and liver tissue SOD, ADH and ALDH activity, increase GSH content, reduce the content of MDA and alcohol, in order to the role of fast sober up.4. The protective effect of S-u on the liver tissue. The rats were treated with S-u, physiological saline and "Ganlixin"(diammonium glycyrrihizinate) for14days, and then were gavage with "Erguotou"(20ml/kg). After3h, the eyeball blood was collected and transformed into serum by refrigerated centrifuge. The alanineaminotransferase (ALT), aspartate aminotransferase (AST), catalase (CAT), SODactivity, GSH and MDA were detected. It was found that the S-u might reduce ALT, AST significantly and improve the activities of SOD, CAT et al. Thus it has a better protective effect on acute alcoholic liver injury. |
