Toxoplasma gondii is a widely parasitic human and many kinds of mammals,opportunistic and obligate intracellular parasitic protozoa, it can be parasitic allnucleated cells in addition to the red blood cells in the hosts, and cause serious zoonosisof toxoplasmosis. When the human body infection with T. gondii, the normal immunefunction shows asymptomatic of recessive infection, but the Immunodeficiency or thehandicapped patients infection with T. gondii will leads to the deterioration of thepatient condition, prognosis is poor and even death. Human immunodeficiencyvirus(HIV)is a kind of slow virus infection of human immune system cells, the HIVmainly affects the body’s immune system after enters the body, attack and destruction ofCD4+T helper cells and macrophage system. Damage to the person’s immunity, leadingto extreme low immunity. Thus secondary all kinds of serious opportunistic infectionsand tumors, eventually cause patient death. As the HIV infection rate increased year byyear, the world has been paid more attention to the diseases related T. gondii infection.Yunnan province is China’s multi-ethnic frontier provinces, its special geographicallocation (near the world’s most famous drug distribution center "golden triangle"), isone of the provinces that China’s earliest report human HIV infections and have alarge number of infected persons at present. We did the study,in order to understand T.gondii infection situation in the HIV positive population in Yunnan province and thebody’s immune cells reaction after T. gondii infection.1. Objective1.1. Preliminary understanding T. gondii infection prevalence in HIV positivepeople of different geographical and ethnic, gender, age in western Yunnan.1.2. A preliminary study of the body’s cellular immune gamma interferon, interleukin-2, interleukin10, nitric oxide(IFN-gamma, IL-2and IL-10, NO) thedynamic changes of the concentration after HIV positive people infect with toxoplasma.Understanding situation of immune function in HIV positive people infect withtoxoplasma.2.MethodsAccording to the AIDS epidemic in western region of Yunnan,1839serumsamples of HIV positive have collected from Dali, Baoshan, Dehong and Lincang4states (city) in Yunnan province. Another collection of local HIV negative peopleserum samples180.Detect IgG/IgM antibody levels of T. gondii all of serum samples collectedHIV-positive patients and to HIV negative people by use polymerase Enzyme leagueimmune (enzyme-linked immunosorbent assay, ELISA). After the first detection, the T.gondii IgG/IgM antibody positive patients follow-up about six months, collect the bloodsampling Once again and test the concentration of IL-10, IFN-γ, IL–2, NO withELISA. And comparison with HIV negative people have no T. gondii infection.3. Results3.1T. gondii specific antibody IgG, IgM positive rate is HIV positive people were35.3%,1.8%, In the western region of Yunnan, Dali prefecture, Baoshan city, Dehongprefecture and Lincang city IgG positive rate were30.9%,33%,41.9%,40.6%, IgMpositive rate were1.1%,2.2%,3.0%,2.3%, statistics analysis have differencesignificant (p﹤0.05). T. gondii antibody IgG, IgM positive in HIV negative people ratewas23.3%,1.1%.3.2The test results of T. gondii IgG, IgM antibody in HIV positive people betweendifferent national: T. gondii IgG, IgM antibody positive rate in the Jingpo nationality isthe highest56.7%4.1%, the hui nationality were the lowest20.6%and0%, Positiverate of T. gondii IgG have significant differences between different ethnicgroups(p<0.05), IgM positive rate had no significant difference(p﹥0.05).3.3T. gondii IgG, IgM antibody test results of different gender HIV positivepeople: male36.1%,1.9%, female34.2%,1.8%, T. gondii IgG and IgM infection rate between male and female have no significant difference (p>0.05).3.4The test results of T. gondii IgG, IgM antibody in HIV positive people betweendifferent age:41~60years group T. gondii IgG antibody positive rate is the highest38.1%,﹤20years group is the lowest21.1%,>60groups IgM positive rate is thehighest3.2%,21-40group is the lowest1.6%, T. gondii IgG, IgM, infection ratesbetween different age have no significant difference (p﹥0.05).3.5Concentration of IL-10(178.54±60.23) after the follow-up is significantlylower than before the follow-up (201.23±57.96) in HIV positive people that T. gondiiIgG antibody positive (p﹤0.05). Concentration of IFN-γ、 IL-2after thefollow-up(436.76±213.76,292.59±162.81) is significantly higher than before thefollow-up(376.79±199.66,248.73±143.89),(p﹤0.05)、(p﹤0.05). Concentration of NO(84.18±78.15) is significantly lower than after the follow-up (81.37±77.58),(p>0.05)The concentration of IL-10, IFN-γ, IL-2, NO after follow-up of is no significantdifference between before and after the follow-up in T. gondii IgG and IgM antibodydouble positive people (p>0.05). There was no significant difference between beforeand after the follow-up concentration of IL-10, IFN-γ, IL-2, NO in HIV positivepeople that T. gondii antibody negative(p>0.05). Dually infected with HIV and T.gondii test first and second concentration of IFN-γ、IL-2are less than people single withHIV and IL-10、NO are higher than people single with HIV.4. Conclusion4.1The infection rate of T.gondii with HIV positive people were higher in west ofYunnan, relating to the regions and habitat of nationalities.4.2The cytokines that secreted from Th1cells were the main ingredients during theperiod of chronic T.gondii, which can produce anti-infectious immunity.4.3The acute infections which were caused by recurrence or newly infected withtoxoplasma, cellular immunity of patients can be subject to a certain degree ofinhibition.4.4HIV positive people who infected toxoplasma would accelerate the damage of bodyimmune function. NO played a part in killing intracellular toxoplasma and restraining the growth of the worm propagation in the infection of toxoplasma.
