ObjectiveAlong with the humanity economic society development, China hasentered into the aging society period. the health of old folks alsobecomes more and more important.It is now an urgent question toexperiment how to delaying senescence,especially for learning andmemory abilities decline due to aging.it badly affects people’sliving standards. Acupuncture has characteristics and advantagesin anti—aging aspect. This study adopt Shuangguyitongelectroacupuncture therapy, to investigate the influence onlearning and memory in aging rats.Furthermore,it provides a certainexperiment basis on electroacupuncture regularion of aging.Toexplore the underlying mechanism of electroacupuncture foranti-aging.MethodsThirty there-month-od SD rat(male),ten of them were randomlyselected into normal control group(NC),aging model group(model),shuangguyitong electroacupuncture treatment group(EA),10rats ineach group.Aging model(in the late2groups except the controlgroup)was duplicated by subcutaneous injection of D-galactose inthe back skin with a calculate of12mg/kg.d for40days,other ratsin normal control group were injected with the same way but the substance is stead of D-galactose.The successful model accountfor.The rats of electroacupuncture treatment group wereacupunctured on Guanyuan(GV4)﹑Housanli(ST36)﹑Baihui(GV20) inwhich refer to the book of《experomental acupuncture》editor by LiZhongren.The needle was25mm in the length and0.35mm in theDiameter Huatuo license,and HANSLH202H electro acupuncture therapyapparatus.We pick up Guanyuan and Housanli by a pair ofelectrodes,and bilateral Zusanli is in alternate use of the nextday.We selected a continuous-wave,frequency of2Hz, currentintensity of lmA,and treated for15minutes,once every day,6timesa week,lasting four weeks.After the end of treatment,we tested in rata by Morris watermaze in order to detection the learning and memory abilities of ratsfor five days.The rats were sacrificed an the end of the second dayafter taking the hippocampal tissue,then the tissue is in SABCimmunohistochemical assay detection of the level of expression ofCalmodulin,CaM,with Western blot assay we select the receptor ofCa2+、Calcium/calmodulin-dependent protein Kinase Ⅱ,CaMKⅡ andCyclic AMP Response Element-Binding, CREB.ResultsCompared with the normal controlled group,the activity ofhippoampal CaM、CMKⅡ、CREB protein content were lower and theabilitiy of learning and memory declines dramaticaly in the agingmodel group.(P<0.01).Compared with the aging model group, the activity of hippoampalCaM、CMKⅡ、CREB protein content were higher and the abilitiy oflearning and memory significantly improved in the shuangguyitongelectroacupuncture treatment group.(P<0.05). ConclusionThe abilitiy of learning and memory declines dramaticaly in theaging model group compared with the normal controlled group,itsactivity of hippoampal CaM、CMKⅡ、CREB protein content were lower,probably is one of the mechaniams that lead to the reduction of thelearning and memory ability. The abilitiy of aging model’s learningand memory significantly improved after the treatment ofshuangguyitong electroacupuncture,the activity of hippoampal CaM、CMKⅡ、CREB protein content were higher.This suggests that thetreatment of shuangguyitong electroacupuncture may improve theabilitiy of aging model’s learning and memory by regulation thesignaling pathway of hippocampal tissue CaM-CaMKⅡ-CREB.
