Objective Parkinson’s disease(Parkinson’s diseases,PD)is acommon nervous system degenerative disease in middle-agedand old. Main pathological changes in the substantia nigra andstriatum pathway,dopamine due to the decreased production,leadto increased static tremor,muscle tension,bradykinesia,etc.Chineseand western medicine treatment of Parkinson’s disease history canbe traced back to1867,respectively,and the han and tangdynasties period in our country.Western medicine treatmentexperience from belladonna alkaloid dopamine to landmarkdevelopment,the modern understanding of PD of syndromedifferentiation and treatment experience of traditional Chinesemedicine is also a trend of deepening[1].But mostly adoptmedication and represented by surgery damaged nuclear group ofsymptomatic treatment. But above treatment did not change theloss of dopaminergic neurons within the environment,can not stopthe continued loss of neurons make disease has progressivedevelopment, thus can not achieve satisfactory long-term effect,and clinical exist large dose of levodopa preparations madeobvious deterioration,the condition of high incidence of long-termmedication side effects[2].Stereotactic surgery also have intracranial hemorrhage,facial paralysis,dysarthria caused by theoccurrence of complications such as[3].This experiment throughthe detection of ubiquitin,a alpha-synuclein(a-synuclein,Syn),ubiquitin activating enzyme(E1),ubiquitin ligase(Parkin),ubiquitin-carboxyl terminal hydrolase L1(UCH-L1)content,observation model rat substantia nigra of Parkinson’s disease areain the ubiquitin-proteasome pathway of multiple molecularexpression and the curative effect,get the basic data,Parkinson’sdisease model rat substantia nigra dopaminergic neurons functionof ubiquitin-proteasome system activity,protein precipitationconditions and the effect of acupuncture therapy on the system,observe electricity for Parkinson’s disease model rats ubiquitin-proteasome system function,the influence of the ubiquitin-proteasome system was revealed in the role of acupuncturetreatment of Parkinson’s disease,to explore the curative effect ofacupuncture therapy for Parkinson’s disease and the possiblemechanism. From the ubiquitin-proteasome system angle toexplore the mechanism of electroacupuncture therapy onParkinson’s disease(PD).Methods50rats were randomly divided into5groups: a normalgroup, a dissolvent contrast group, a model group,a pretreatmentgroup, a electroacupuncture therapy group;The Parkinsonian ratmodel was built by hypodermic injection of rotenone.While therats’ behavioral changes were observed, western-blottingmethod was adopted to detect parkin, UCH-L1,UBE1expression inthe regions of substantia nigra, immunohistochemical stainingmethod was used to observe a-synuclein, ubiquitin and THpositive cells quantity. Results The rats of model group showed some characteristicchanges, such as yellow and rough hair, arch back, weakness ofescaping arrest,slowing movement,while the abnormal changes ofelectroacupuncture therapy group and pretreatment group weresignificantly slight. The Parkin, UCH-L1, UBE1, UB and THexpression of model group rats significantly decreased, buta-synuclein expression significantly increased (p<0.01). Suchabove indices could be partially reversed by the intervention ofpretreatment or electroacupuncture therapy (p<0.01).Conclusion Ubiquitin-proteasome dysfunction may be involvedin the pathogenesis of Parkinson’s disease,electroacupuncturetreatment or pre-treatment may have a protective effect on ratmodel of Parkinson’s disease,and enhance theubiquitin-proteasome function,suggesting that acupunctureprevention and treatment of Parkinson’s disease couldmechanically attribute to improving the function ofubiquitin-proteasome system.
