Objective:To study the regulation of miRNA-92a on TrkA protein expression andits effect on the proliferation and migration of human neuroblastoma cells.Methods: The expression of miRNA-92a and TrkA mRNA were quantitativelydetected by RT-qPCR before and after transfected with miRNA-92a mimics ormiRNA-92a inhibitor. The proliferation and migration of neuroblastoma cells wereexamined in vitro by CCK-8and transwell assay. The expression of TrkA protein wasquantitatively detected by Flow Cytometry.Results:1. RT-qPCR analysis showed that compared with negative control, themiRNA-92a expression was increased after miRNA mimics transfection, while themiRNA-92a expression was decreased24hours after miRNA inhibitor transfection.2.CCK-8assay and direct cell count showed that compared with negative control, theproliferation of neuroblastoma cells in group of miRNA-92a mimics transfection wassignificantly increased while it was significantly inhibited in the group of miRNA-92ainhibitor transfection group.3. Transwell migration assay resulted that the number ofcells across the membrane in group of miRNA mimics transfection was increasedcompared with that in control group. The number of cells across the membrance ingroup of miRNA inhibitor transfection was decreased compared with that in controlgroup.4. RT-qPCR analysis of TrkA mRNA showed that compared with negtive control,the TrkA mRNA expression in group of miRNA-92a mimics transfection wassignificantly decreased while the TrkA mRNA in group of miRNA-92a inhibitortransfection was significantly increased.5. Flow Cytometry assay for TrkA proteinexpression showed that compared with negtive control, the TrkA protein expression ingroup of miRNA-92a mimics transfection was significantly decreased while the TrkAprotein in group of miRNA-92a inhibitor transfection was significantly increased.Conclusion: miRNA-92a may down regulate the TrkA mRNA and protein expression and promote proliferation and migration of human neuroblastomaBE(2)-M17cells.
