BACKGROUNDLiver cancer is one of the most common malignant tumor in clinical practice.Hepatocellular carcinoma is the most common type of liver cancer, accounting for70%~80%oftotal cases. In order to further study the pathogenesis of liver cancer, to explore new methods oftreatment, it is crucial for experiment to establish a scientific animal models of liver cancer.Through comparing the difference of tumor incidence and tumor characteristics betweenHep G2cells and Bel-7402cells under different conditions, we want to establish a suitableanimal model of liver cancer for further experiments.OBJECTIVE1. To study and compare the difference of tumor incidence and incubation period betweenHep G2cells and Bel-7402cells under different conditions.2. To study and compare the difference of tumor growth rate between Hep G2cells andBel-7402cells, and do an induction of tumor growth rule.3. To study and compare the difference of morphological features between Hep G2cellsand Bel-7402cells. To detect serum AFP concentration and perform a pathological andimmunohistochemical analysis.METHODS1.90BALB/c nude mice were randomly divided into9groups. According to different cellconcentration and age of mice, Hep G2cells and Bel-7402cells were injected into micesubcutaneously, with9th group as a blank control group. Then to study and compare thedifference of tumor incidence and incubation period between Hep G2cells and Bel-7402cellsunder different conditions.2.To detect the size of tumors with vernier caliper weekly. The calculation formula oftumor volume is V=π ab2/6(V: volume, a: long diameter, b: short diameter).3. To detect the serum AFP concentration of tumor-bearing mice with the chemiluminescence method. To study and compare the difference of morphological featuresbetween Hep G2cells and Bel-7402cells. Finally the postoperative specimens were performedfor histopathological and immunohistochemical examinations.RESULTS1. When cell suspension concentrations of Hep G2cells reached5.0x107/ml, we can gettumor model successfully not only in younger mice group(4~8week age) but also in older micegroup(8~12week age). As for the Bel-7402cells, when the cell suspension concentrationsreached5.0x107/ml and was injected subcutaneously in younger mice(4~8week age), then wecan obtain a higher tumor incidence.2.There were no same incubation period in two kinds of hepatoma cells under the sameconditions. The incubation period in Hep G2cells was19.4±5.2days; while in Bel-7402cells itwas34.3±15.7days.3. Subcutaneous tumor established by Hep G2cells was composed of multiple nodules ofwhite sample mass. While the center of subcutaneous tumor established by Bel-7402cells wasnecrotic tissue.4. Two kinds of cells were negative AFP expression. And the detection of Hep Par1byimmunohistochemical method was positive in tumor of Hep G2cells but negative in tumor ofBel-7402cells.CONCLUSION1. Hep G2cells are more likely to form a subcutaneous tumor in nude mice than Bel-7402cells.2. Hep G2cells has a shorter incubation period and grow faster than Bel-7402cells.3. Two kinds of cells are negative AFP expression.
