Objective:To observe hypercholesterolemia on injury of rabbits myocardialmitochondria,To study the presence protective effect of atorvastatin and CoQ10.Methods:40two moons aged healthy male New-Zealand albino rabbits were used. Theseexperiment rabbits were randomly divided into control group(A group);high fat dietgroup(B group);high fat diet+atorvastatin group(C group);high fat diet+atorvastatin+low dose CoQ10group(D group);high fat diet+atorvastatin+high doseCoQ10group(E group).All rabitts were sacrificed6weeks later,removed the heartquickly, their fresh tissue of heart apex were taken and prepared to myocardialmitochondria and electronmicroscopy specimen.To detection the content of MDA andGSH in myocardial mitochondria in each group and to observe the myocardialmitochondrial morphological changes by electronmicroscopy.Results:Comparing withA group and E group the content of MDA in myocardialmitochondria was increased in B group (P<0.05). Comparing with A group,thecontent of GSH in myocardial mitochondria was decreased in B group (P<0.05).Comparing with E group the content of GSH in myocardial mitochondria wassignificantly decreased in B group (P<0.01).By electron microscopy observation: Comparing with control group, High fat dietgroup myocardial cell ultrastructure injury seriously, part of the fracture,dissolution,mitochondria Obvious swelling,crest structure fuzzy and difficultto recognize,some partial or all of the fault disappear,cause cavitation increase。buthigh fat diet+atorvastatin+high dose CoQ10group myocardial cells ultrastructuremaintain well, close to control group.High fat diet group and high fat diet+atorvastatin+low dose CoQ10group myocardial cell ultrastructure have differentlevels of injury, between the two groups。Conclusion:Hypercholesterolemia can injury rabbits myocardial mitochondria andmyocardial cells;atorvastatin and high dose CoQ10might lessen the injury ofoxygen free radicals in myocardial mitochondria of rabbits by decreasing the contentof Lipid peroxidation (MDA) and increasing the content of antioxidant GSH inmyocardial mitochondrion and protective structure myocardial cell..
