OBJECTIVE:To investigate the effect of2-deoxy-D-glucose (2-DG) on proliferationand apoptosis of human ovarian cancer SKOV-3cellsMETHODS:The human ovarian cancer SKOV-3cells were treated with2-DG indifferent concentrations(2.5mmol/L、5mmol/L、10mmol/L、20mmol/L、40mmol/L).Then MTT was used to detect the cell growth inhibition effect of each group atdifferent time; Hoechst33258staining assay was chosed to observe the morphologicalchanges induced by2-DG; the cell cycle and cell apoptosis were analyzed via flowcytometry.RESULTS: Different concentration of2-DG can inhibit the proliferation of SKOV-3cells in a dose and time-dependent manner, compared with control group, and there issignificantly difference (P <0.05);Our results also showed2-DG treated cells were arrestedin G0/G1phase; Apoptosis assay by Flow cytometry admonsted that2-DG can induce cellapoptosis or necrosis.CONCLUSION:2-DG has inhibitory effect on the proliferation of SKOV-3cells invitro, and the effect is dose and time-dependent.
