Objective:This study aims to establish the CPR model in rabbits in order to demonstrate the neuroprotective effects of ethanol after post-cardiac arrest.Methods:40rabbits were randomly divided into five groups:sham group, the control group,0.5g/kg group,1.0g/kg group, and1.5g/kg group. After the establishment of CPR model, those rabbits were injected with different doses of ethanol immediately following the restoration of spontaneous circulation. After2h,8h and12h, S-100protein expression were tested respectively in order to determine the dose-response effect of ethanol in brain necrosis volume.Results:1The brain necrosis volume of different groups. The brain necrosis volume of the sham group, the control group, the0.5g/kg ethanol group, the1.0g/kg ethanol group, and the1.5g/kg ethanol group are5.26±0.26,29.2±1.46%,25.0±1.25%,20.4±1.02%, and15.3±0.76%respectively. Among the four groups,1.5g/kg ethanol group has the most significant neuroprotective effect (p<0.001)2Serum S-100concentration of the different groups after2h,8h and12h. After2h, Serum S-100concentration of the sham group, the0.5g/kg group, the1.0g/kg group, the1.5g/kg group, and the control group are1.19±0.05μg/L,1.04±0.05μg/L,0.35±0.01μg/L,2.25±0.11μg/L and0.06±0.03μg/L respectively. S-100concentration of the1.5g/k group is the lowest one (p<0.001) After8h, S-100concentration of the0.5g/kg group, the1.0g/kg group, the1.5g/kg group, the control groupand the sham group, are3.96±0.19μg/L,2.12±0.10μg/L,0.45±0.02μg/L, and4.03±0.20μg/L and0.06±0.06μg/L respectively. The S-100concentration of the1.5g/kg group is lowest one (p<0.001) After12h, the S-100concentration of the0.5g/kg group, the1.0g/kg group, the1.5g/kg group, the control group and the sham group are5.58±0.27μg/L,496±0.24μg/L,0.98±0.04μg/L,7.25±0.36μg/L and0.06±0.01μg/L respectively. The serum S-100concentration of the1.5g/kg group is the lowest one (p<0.001) Conclusion:The result of the experiment suggests that an immediate injection of a dose of1.5g/kg ethanol in rabbits immediately following the restoration of spontaneous circulation have neuroprotective effect.
