Objective:Case-control study method was adopted to explore the pre-miRNA gene polymorphism in Chi nese han populations of the correlation between chronic obstructive pulmonary disease. Including the pre-miRNA genes:rs2910164G/C,rs3746444C/T,rsl1614913C/T.METHODS:In the first hospital of Lanzhou university and West China hospital of sichuan university respiratory screening of clinical diagnosis of532cases without blood relationship clear COPD patients and511cases of normal control group. Collection of COPD patients and normal control group blood samples, centrifugal column method is used to extract total genomic DNA samples, using UV spectrophotometer instrument measuring the concentration of DNA, and then design the site specific amplification primer, using polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) method to analysis three SNP loci located in the pre-the miRNA gene (rsl1614913, rs3746444and rs2910164) in Chinese han population in COPD patients and normal allele and genotype frequencies, use support SPSS11.5statistical software to analyze the differencesResults:The rsl1614913C/T and rs3746444A/G locus allele frequencies and genotype frequencies distrib ution between case group and control group show significant difference. The rsll614913C/T polymorphi sms of the CC genotype and C allele and was significantly associated with an increased incidence of C OPD(CC contrast TT, OR=0.55,95%CI,0.372-0.818; C对比T, OR=0.806,95%CI,0.676-0.962). S imilarly, rs3746444A/G polymorphisms of the AA genotype and allele A also showed a strong correlatio n with increased incidence of COPD(AA contrast GG, OR=0.158,95%CI,0.046-0.542; A对比G, O R=0.733,95%CI,0.569-0.943).Conclusion:Research shows that rsl1614913C/T and rs3746444A/G polymorphism in the process play a n important role in the pathogenesis of COPD, prompt the pre-micrornas hsa-mir-196-a and has-mir-499gene in the han population of northwest and the risk of developing COPD has a strong correlation, b ut the pre-micrornas hsa-mir-146-a did not show the correlation of COPD disease with the han Chinese population.
