BackgroundAt present, the incidence of lung cancer increased, the mortality rate of malignant tumors is in the first place. Non small cell lung cancer (NSCLC) accounts for about85%of lung cancer, in which about70%of the patients have been diagnosed in advanced stage. Chemotherapy is the main curing method, and the standard chemotherap is two solution containing platinum. The efficiency is only about25%-35%, the median survival period of8-10months, has entered a bottleneck period. In recent years, molecular targeted therapy for certain types of cancer (such as EGFR mutation, EML4-ALK positive) shows better result, but these drugs will sooner or later got resistance, how to overcome the resistance and find new targets and new drug become the focus of clinical research.ObjectivesThrough the detection of epidermal growth factor receptors (EGFR) and hepatocyte growth factor receptor (c-Met) expression levels in non-small cell lung cancer (NSCLC) tissues, we try to figure out the relationship between c-Met and EGFR protein expression and the histological type and clinical stage.Method 60cancer tissues of NSCLC patients who got the surgery in the Second Affiliated Hospital of Zhengzhou University during2010.2and2011.8. were collected, and the EGFR and c-met protein level were detected by immunohistochemistry and hybridization assay in these tissues.We Use SPSS17.0statistical analysis software and x2test (test level is set to a=0.05) to analyze the relationship among immunohistochemistry results of EGFR and c-Met, histological type and clinical stage.Results1.EGFR protein expression rate was80%(28/35) in adenocarcinoma of lung EGFR protein expression rate was12%(3/25) in lung squamous cell carcinoma, the expression of EGFR protein in adenocarcinoma was significantly higher than that in squamous cell carcinoma (P<0.05), the positive expression rate of c-Met is62%(22/35) in lung adenocarcinoma, and it is40%(10/25) in lung squamous cell carcinoma, the expression of c-met protein in adenocarcinoma was significantly higher than that in squamous cell carcinoma (P<0.05).2.Through Logistic analysis, we found that the influencing factors of EGFR protein expression were gender, histological type, lymph node metastasis and TNM stage (P<0.01), the influencing factors of the c-Met protein expression were the differentiation degree, tumor size, histological type, lymph node metastasis and TNM staging condition (P<0.01). Expression of both EGFR and c-Met in three and four stage patients was higher than that patients in stage one and two(P<0.01).3.The positive rate of EGFR and c-Met expression in one and two stage of NSCLC tissues was18%(16/34), three and four stage was34.5%(15/26), there was significant difference between the two groups of positive expression4. There is No correlation between the expression of c-Met and EGFR (rs=0.341, P>0.005).Conclusions1.The expression of EGFR and c-Met protein in lung adenocarcinoma was higher than in squamous;2.Expression of EGFR and c-Met in stage three and four was significantly higher than that in stage one and two. 3. The higher EGFR and c-Met double positive expression, The higher malignant degree of tumor tissue.
