BackgroundHepatocellular carcinoma is a common gastrointestinal cancer, the recurrence and metastasis is a major obstacle to treatment of liver cancer. In recent years, studies have shown that filamin A (filamin A, FLNa) is an important scaffold protein in cellular signal transduction,and it involved in proliferation, adhesion, migration and other cell behavior,FLNa is closely related with tumor invasiveness, and impacts with the occurrence and development of tumors. So we Suggest that FLNa may play an important role in the progression of liver cancer.ObjectiveTo investigate the relationship between FLNa expression and the clinicopathologic characteristics of the hepatocellular carcinoma by detecting the expression of FLNa in the peripheral blood and tissues of patients with hepatocellular carcinoma,so as to provide new ideas for liver cancer research.MethodsCollecting156patients’serum and tissue samples,and156patients divided into liver cancer group(97cases) and normal control group(59cases), normal control group includs liver hemangioma of47cases, hepatolithiasis in7cases,5cases of liver injury; with ELISA method to measure serum concentration of FLNa,and the expression of FLNa in tissues were analyzed by immunohistochemical method.ResultsIn the serum,the average value of FLNa in the patients with hepatocellular carcinoma and normal control group was31.64±10.51pg/ml and47.16±18.62pg/ml repectively, the FLNa expression in hepatocellular carcinoma was significantly lower than that in normal control group(P<0.05). In the tissues, the positive rate of FLNa expression in liver cancer group was21.64%, and83.05%in normal control group, the expression of FLNa in the liver cancer group was also lower than that in normal control group(P<0.05). In the meantime,the patients who have the higher AFP value, lager tumor size, low differentiation degree,and have happened relapse and metastasis, the expression of FLNa in the serum and tissue was significantly lower (P<0.05), but there is no significant difference in the different sex and ages of the patients.Conclusions1.The expression of FLNa in the serum and tissue of liver cancer group was significantly lower than that in normal control group, and the low expression of FLNa may be related to the differentiation and invasiveness of the liver cancer.2.Detection of the serum concentration of FLNa can be used to diagnose hepatocarcinoma and predict the progression of liver cancer, and with further study in FLNa, may provide new ideas for the diagnosis and treatment of liver cancer.
