Diabetes is a common and frequently-occurring disease harm people’s health, with the change of the human living environment and way of life, the number of diabetics in the world having a sharp increase, diabetes has become the third largest disease after cardiovascular disease and cancer, which in the21st century became a danger killer to human health. How to prevention and treatment of diabetes is one of the major research topics within the scope of the world, research and development for the treatment of diabetes drugs are attaches great importance to human beings. Current clinical treatment of diabetes is mainly rely on injections of insulin and oral hypoglycemic drugs, but its many side effects, and high treatment cost, and bring heavy financial burden. The traditional Chinese medicine for the treatment of diabetes in our country has a long history, and its effect is remarkable. So, research and development of glucose-lowering drugs which have efficient and small side effects from the traditional Chinese medicine has become a hot topic. RM is regarded as a commonly used traditional Chinese medicine for the treatment of diabetes. Modern research has shown that RM have the function of lipid-lowering, step-down and increasing the body’s endurance, anti-aging, lowering cholesterol, inhibiting thrombosis, inhibiting harmful intestinal bacteria breeding, inhibiting harmful oxide formation, inhibiting cancer and adjustment of adrenalin function effect, but the most prominent feature is the ease of diabetes. This thesis as α-glucosidase for targets to research in vitro,and normal mice and diabetic mice induced by the STZ as the research object in the body, to search for the hypoglycemic activity on extract of RM, and the main content of the research are:(1). This study sift the higher activity parts as α-glucosidase for the targets through different of the water extract of Cortex Mori(CM), Mulberry Leaf(LM) and RM, and come to the conclusion that three parts both have the inhibition of α-glucosidase. Comparison among them, Cortex Mori has the highest inhibition rate (IC50=0.27mg/ml), then RM, and LM is a bit poor. In the early animal experiments, three parts also can reduce the mice (normal or diabetes) postprandial blood glucose (PBG) levels. In combination with other factors, such as a large number of RM will be aborted on November each year, RM resources are rich, and it seems to be easy to implement on the production scale when compared with CM. So adopted RM is the raw materials to study in the laboratory.(2). This study established the process on laboratory level of extracting total alkaloid, total sugar and total flavonoids from RM, as α-glucosidase for the targets, three different kind of components and its monomer was prepared from the crude drugs of RM by using macroporous resin, alcohol precipitation, silica gel adsorption and recrystallization and so on, then the main chemical composition of the three extracts were analyzed by chemical analysis,three kinds of main components were alkaloids, polysaccharides and flavones.(3). Three kinds of active components and its representative monomer (polysaccharide, DNJ, rutin) from the water extract of RM be tested the role for α-glucosidase in vitro, the result shows that the inhibition ability of α-glucosidase for alkaloids (DNJ)> flavonoids (rutin)> polysaccharides (polysaccharide), especially the inhibition activity of DNJ is very strong while polysaccharides is very weak. Each monomer compounds both can reduce the PBG of normal mice in the body levels, and delay the time of peak of blood sugar. After30min, drug groups compared with blank control group, there were significant differences in blood sugar levels (P<0.05or P<0.01); then study the effect of PBG by monomer compounds on experimental diabetic mice, when drug groups compared with blank control group, the PBG values of DNJ group significantly decreased (P<0.01), rutin and polysaccharide group have significant difference (P<0.05).(4).The Compound ingredients which contain the main hypoglycemic active ingredient DNJ,rutin and polysaccharide in vitro enzyme level, the inhibition effect of α-glucosidase were superior to single components. Glucose tolerance experiment in normal mice and experimental diabetes mice, after30min dose, Compound group compared with DNJ group, PBG values have significant difference (P<0.05), proved that DNJ, rutin and polysaccharides have a good synergistic effect when blend with each other.
