Objective Thisissue through injected6-hydroxydopamine(6-hydroxydopamine,6-OH--DA)into striatum to established a Parkinson’s disease animal model,electroacupuncture "Baihui> Fengfu" point, observed EA for its behavior, the substantia nigra dopaminergic neurons in the number of apoptotic and substantia nigra of caspase (cysteinyl aspartate specific proteinase)-8and caspase-3’s expression. In this experiment, we are starting from the angle of apoptosis occurred angle,focusing on the role of caspase-3and caspase-8in Parkinson’s disease of rat model substantia nigra dopaminergic neurons during apoptosis,and the electroacupuncture on its impact,to further discuss the mechanism of acupuncture treatment for Parkinson’s disease,and to provide experimental evidence for the clinical application of acupuncture in the treatment of Parkinson’s disease.Methods healthy male Sprague Dawley rats were Selected85,weighted180-220g. Those rats were randomly divided into Sham Group,Model Group,Electro-acupun cture Group,then according to the treatment time each group was divided into three subgroups of2weeks Group,3weeks Group and4week Group.The research created rats model of PD which caused by injecting6-hydroxydopamine into the left striatum from4points use stereotaxic apparatus.The sham operation group was injected with the same amount of saline which containing ascorbic acid. After7days,apomorphine was injected into the rats neck subcutaneously to induce rotation.This test was repeated every7days.Those who had constant right rotation with a speed faster than7revolutions per minute were selected as successful PD models.Were treated for2weeks,3weeks and4weeks after, the rotation induced by the subcutaneous injection of APO (0.5mg/kg), five minutes after the injection began to record the number of rotations of the rats to the contralateral constant direction, recorded a total of30minutes to detect EA affect on it’s behavior. By Nissl staining and terminal-deoxynucleotidy transferase mediated nick end labeling(TUNEL) method observed the morphologic change of substantia nigra neuron and the neuronal apoptosis in substantia nigra in each group. Histochemical and immunohistochemical staining methods were adopted to observe the expression of caspase-3and caspase-8in substantia nigra in each group.Results①the affect of Parkinson’s disease rat model’s behavior by EA; Compared with the same period model group, the number of rotations were significantly reduced in3and4weeks EA group (T<0.05,.P<0.01),the number of rotations in2weeks EA group had little difference(P>0.05); Compared with2weeks EA group, the reduce of the rotations were significant differences in3,4weeks EA group (P<0.05,P<0.01),4weeks EA group Compared to3weeks EA group have reduced, but no statistical significance (P>0.05).②the affect of substantia nigra dopaminergic neurons apoptosis on Parkinson’s disease rat model by EA:Compared with the apoptotic quantity in the substantia nigra of same period model group,there were significant difference in3and4weeks EA group (P<0.01), were little reduce in2weeks EA group (P>0.05).③the affect of substantia nigra of caspase-3and caspase-8expression by EA: Compared with the same period model group, the expression of caspase-8and caspase-3in4weeks EA group had significant differences (P<0.01); in3weeks EA group, the expression of caspase-8had significant difference (P<0.01), caspase-3had little difference(P>0.05);the expression of caspase-8and caspase-3in2weeks EA group had little difference(P>0.05).Conclusion①EA intervention can reduce the number of rotations of the model of Parkinson’s disease rat.②EA can inhibit substantia nigra dopaminergic neurons number of apoptotic in the model of Parkinson disease rat.③protein caspase-8and caspase-3expression in model group were increased, EA group the expression was inhibited.
