Objective: This study was designed to investigate the effects of leadexposure on rat cardiovascular system. To observe the molecular mechanismsof proliferation and apoptosis of the cardiac cells and vascular cells during thelead exposed process.Method: We Selected52rats that were health,3-4-week-old and weightin the range of99-114g. SD rats were divided into the normal control groupand the experimental group by random. The experimental group drank contentof1%lead acetate solution, and the control group only drank tap water.After12days, we killed13rats of each group and collected the heart and bloodtissues. After40days we killed13rats of each groups and collected the heartand blood tissues. Extracting protein and nucleoprotein and using western blotto detect the change of proliferation, apoptosis, and the histone acetylation inthe cardiac cells and vascular cells. The western imprinting was scaned by gelimage analysis system. To analyse the result of the western imprinting byFrench Vilber Lourmat company Bio ID digital imaging analysis softwarequantitatively. The data was analysed by T Test, SPSS13.0statisticalsoftware.Results:1The expression of Proliferating Cell Nuclear Antigen (PCNA), B-celllymphoma-2(Bcl-2), Bcl-2Assaciated X protein (Bax) and histone H3acetylation in the cardiac cells with lead exposing for different timeThe results of Western blot and computer western blot imagesemi-quantitative analysis showed that the expression of PCNA of rat heartcells was increased and statistical difference exposed to lead for12days and40days, compared with the normal control group. The results of Western blotand computer western blot image semi-quantitative analysis showed that the expression of Bcl-2and Bax in rat heart cells was increased exposed to leadfor12days. The expression of Bcl-2reduced and the expression of Baxincreased exposed to lead for40days (P <0.05). To compared with thenormal control group, the western blot and computer western blotsemi-quantitative image analysis results show that the level of histone H3acetylation increased in rat cardiac cells exposured to lead for12days and40days.2The change of expression of PCNA, Bcl-2, Bax and histone H3acetylation in aortic vascular tissue with lead exposing for different timeThe results of western blot and computer western blot imagesemi-quantitative analysis showed that the expression of PCNA in aorticvascular smooth muscle cells was reduced, compared with the normal controlgroup (P<0.05) exposed to lead for12days and40days. The results ofwestern blot and computer western blot image semi-quantitative analysisshowed that the expression of Bcl-2and Bax was reduced in the aorticvascular cells compared with the normal control group, in the aortic vascularsmooth muscle cells exposured to lead the expression of Bcl-2reduced and theexpression of Bax increased (P<0.05) exposed to lead for12days. Tocompared with the normal control group, the results of western blot andcomputer westernblot semi-quantitative image analysis show thatthe level ofhistone H3acetylation increased in aortic vascular tissue exposed to lead for12days and40days.Conclusion:1Lead exposure makes the level of heart cells proliferation, apoptosisand histone H3acetylation increase. It suggestd that histone H3acetylationplays an important role in heart cells proliferation and apoptosis.2Lead exposure makes the level of vascular cells proliferationdecreasing and the level of apoptosis of vascular cells and histone H3acetylation increasing. It suggested that the levlel of histone H3acetylationincreases in the process of lead exposure played an important role in vascularcell proliferation and apoptosis.
