Surface enhanced Raman spectroscopy (SERS) can effectively raise optical signal intensity, lower detection limit. Currently, preparation of SERS substrate and application of SERS in biological detection are highly active in SERS research area. In this paper, we focus on the SERS application in immune protein detection.The following studies will be introduced in this paper:1.We choose polymer sphere as template,using in site reduction and growing seeds method to prepared Ag/SiO2hollow microspheres,Ag nanoparticles with50nm radius were deposited on the surface of the PS.Afterwhile we used Polyvinylpyrrolidone as structural directing agents to deposit small particle-formed SiO2to the surface of microsphere. Thus the SiO2particles consists a raspberry shape shell on the surface of the microsphere.Because of the point connection of SiO2particles, large structural holes are distributed on the shell. We further calcine the spheres and remove PS cores to obtain Ag/mSiO2hollow microspheres.2. We investigated SERS signal of Ag/mSiO2hollow microspheres by link the spheres with SERS probe molecule4-ATP. Control different quality of Ag nanoparticles doposit on the surface of PS to see the difference SERS signal between them.Then we choose the best signal substrate for further linked to biological antibodies. At the same time we modified quartz plate with same antibodies too. Then we put quartz plate and hollow spheres into antigen solution. After sandwich like immune reaction, the hollow spheres will be linked to quartz plate as SERS substrate.3. Similarly, after loading~50nm Ag nanoparticles onto the surface of PS core, we modified PS surface with CATB as pore-forming agent. Then we induced TEOS as source of Si and formed SiO2shell on the spheres. We removed PS and CTAB by calvination and obtained mSiO2hollow spheres with Ag nanoparticles inside them. We linked SERS probe molecule4-ATP to hollow sphere and studied the SERS substrate signal of it.
