Phenylboronic acid compounds have attacted more attention just because theirunique physical and chemical properties.Currently, the detection methods based on theunique property of gold nanoparticles(AuNPs) has been extensively applied in proteinã€DNAã€metallic ion and studying the affection between biological molecules. In thispaper, we report a functionalisation assay based on AuNPs for detecting lectin protein.The carboxy group of ithiothymine dipropionate and the amino group of3-amino-phenylboronic acid sulfate directly to happen dehydration by adding1-(3-Dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride(EDC) to form a kind ofmercapto phenylboronic acid derivative(Bor), and the product was characterized byFourier-transform infrared spectroscopy(FT-IR) and matrix-assisted laser desorptionionization time-of flight mass spectrometry(MALDI-TOF-MS).A bifunctional strategy, which enable conjugate phenylboronic acid andpentapeptide CALNN as stable ligand to gold nanoparticles. By using bornic acidscould recognize cis-diol configuration in saccharides in aqueous media, the3’-Sialyllactose(SL) was firstly motified on the gold nanoparticles, which can beemployed to detect three lectins Canavalia ensiformis Agglutinin(ConA)ã€SambucusNigra Agglutinin(SNA) and Maackia Amurensis Lectinâ…¡(MALâ…¡), and analyze theconjugates by UV-visible spectroscopic and transmission electronic microscopic (TEM)techniques. The assay could be used as a optical probes for monitoring specfic affinitiesof lectins with3’-Sialyllactose. |
