In this paper,Ophippholis mirabilis mirabilis from the Changdao Archipelago were used as theexperimantal material.The extraction process of polysaccharide was optimized by the orthogoralexperiment.The protein removaling process,the physical and chemical properties,the sepration andpurification process and part of biological activity,including:the antibacterial activity,the antioxidantactivity and the protection of my ocardical cells were also determined.All of these provide the necessarytheoretical basis for the explotation and utilization of the Ophippholis mirabilis.Orthogonal experiment has been used to investigate the extraction press of the polysaechearide.Theextraction temperature,extraction time,ratio of the material to water and times of extraction were selectedas variable parameters.To creat a multivariate linear model by the orthogonal experiment results.Theresults showed that:the extraction temperature is the biggest factor,the next is the extraction times,the thirdis the extraction time,and the last is the ratio of the material to water.The optimum extraction conditionsfor the experiment is that:the extraction temperature is70℃,the extraction times is four,the extractiontime is4hours and the ratio of the material to water is1:20.The sevag method,enzymatic method and the enzyme-sevag method were used to compare theeffect of the removal of protein.The result showed that the enzyme-sevag method was the best one.Using traditional methods methods to get two polisaccharides from Ophippholis mirabilis,anddetermining part of the conventional physical and chemicial properties,and purification.The improved plate culture counting method was used to determing whethere the sixpolysaccharides have antibacterial activity,and the method was also usde to compare the activity of theantibacterial activity,then the filter paper method was used to cartify the above results.the minimuminhibitory concentration was measured by the improved agar dilution method.The results showed that,allthe polysaccharides had the antibacterial activity.The chemical method was used to determine the antioxodant activity of polysaccharides,includingthe reducing capacity,the scavenging ability of the ultra-cation and the hydroxylion,and the ability of theinhibiting the red cell membrane lipid peroxidation.The result showed that,the polysaccharides of theOphippholis mirabilis had a strong antioxidant capacity.They can affectively eliminate the hydroxylradical,superoxide anin,and inhibit the red cell membrane lipid peroxidation,and its antioxidant capacityand the dose is related. The oxidation model of H9c2was builded by peroxide method.In this paper,the protective effect ofthe polysaccharides for oxygen damage H9c2,apoptosis rate caused by oxygen damage,miochondrialmembrane potential,BC1-2,Bax express,the AKT cell pathway,and the change of Ca2+concentrationcaused by hypoxia were determined.The result showed that the polysaccharides play a role in lowconcentration.
