Development of new drugs are increasingly closely linked to the substances from life itself and the genes and proteins, to understand their structure and the structure of the protein that bears life information is the persistent pursuit of human being. CD has been widely recognized and applied as a tool that can provide important information. CD can make rapid analysis and sensitively detect the subtle changes of protein conformation. The experimental technique is simple, and these advantages make this method known as an important means of researching protein secondary structure.The structure of the protein is rich and varied; the prevailing method of spectral-based analysis of the secondary structure of protein is simple classification for protein structures. The length of the helix, the chain length of the protein and the number of chain will affect the value of the ellipticity. The experimental conditions of CD spectra impact on experimental results. The data in the protein database are difficult to be reproduced in the laboratory as well. The data processing methods are all likely to produce data errors, especially the impact of data fitting. What is most lacking in the research on using the CD spectra to predict half content of the protein secondary structure is a great amount of protein and peptide curve library. The shortage of these standard data has seriously affected the credibility of CD spectra forecasting.This experiment will explore factors affecting protein secondary structure prediction by CD from three aspects:protein sample, experimental conditions and forecast calculation. The measured protein samples, such as lysozyme, ribonuclease ect.,are all proteins first publicly reported and with relatively definite secondary structure, and also exist in PDB.Reference test is applied by FTIR and X-ray standard data is used as reference data.This study aims to draw attention from researchers on the CD spectra to protein secondary structure. In conclusion, the CD spectrum is not a suitable method for estimating the protein secondary structure percentage.The accuracy of this method is the worst. The CD spectrum is easy to be influence by the environmental factors and the reference protein data are insufficient. The CD spectra to estimate protein secondary structure is a wrong application method.
