Salt stress is one of the enviromromental factors that have an influence on thegrowth and development of plants. That the beet has strong salt tolerance is reflectedin that the beet has stronger ability to absorb or hold Na+than other plants.Protein is the basic unit that organism makes its physiological function work. Andit embodys the metabolic activities of life directly. To analyze and appraise its functioncan give a great push to clarify the gene function and can reveal life phenomenon moreobjectively and accurately.In this study, salt-resisting T510strains and salt-sensitive210strains of beet as themain subject, by optimizing the extraction of Beet leaves’ total protein andelectrophoresis conditions, we can ultimately gain high-resolution, reproducible2-DEmaps of salt-resisting T510strains and salt-sensitive210strains. By softwarecomparing two strains280mmol/LNaCl stress group with control group, there are607protein spots that match T510and317for210. By the calculation of the protein spots,there are15protein spots in the T510changed because they are affected by saltstress,8protein spots of express abundance in the salt stress treatments wereweakened, meanwhile7protein spots of express abundance in the salt stress treatmentswere strengthened, two differentially expressed protein spots of them identified asunknown protein; there are12protein spots in the210strains changed because they areaffected by salt stress. Compared with the control group,3protein spots of expressabundance in the salt stress treatments are weakened,8are strengthened., onedifferentially expressed protein spots of them identified as unknown protein, twodifferentially expressed protein spots of them identified two types of protein(P<0.05).By analysising the two dimensional gel electrophoresis of two strains of salt stress, wefind288protein spots can be matched and3can’t (points5and13in210strains, point 11in T510strains).Point5in210strains was down-regulated, meanwhilecorresponding protein spots was up-regulated in T510strains. Point13in210strainswas down-regulated, but lost in T510strains. Point11in T510strains wasdown-regulated, meanwhile corresponding protein spots was up-regulated in210strains.After28differentially expressed protein spots were deducted, digested andMALDI-TOF/TOF spectrometry identified, one point was not identifiedsuccessfully(210strain,point13), the other points were identified successfully.
