Part One Establishment of rat models of pulmonary hypertension induced by monocrotaline[Objective] The aim of our study was to establish the rat models of pulmonary hypertension induced by monocrotaline.[Methods] Eighty Sprague-Dawley rats were randomly assigned into two groups:control group, and monocrotaline (MCT) group (60mg/kg). Rats in PH groups received single subcutaneous injection of MCT. We measured pulmonary artery pressure by right heart catheterization using an improved hand-made PE-50catheter. Cardiac output was calculated through thermodilution method. After injection of MCT, We using MRI to measure the Ventricular Function of the rats every week, synchronously measured by right heart catheterization.[Results] Injection of monocrotaline after two weeks, the right atrium and mean pulmonary artery pressure between rats in PH and the control group showed no significant difference[the first week (23.30±2.50) mm Hg (1mmHg=0.133kPa) vs (21.80±2.74) mm Hg, P>0.05; the second week (25.40±3.78)mm Hg vs(22.40±2.76)mm Hg, P>0.05]; the cardiac output between rats in PH and the control group was no significant difference [the first week (156.49±15.52) ml/min vs (156.49±15.52) ml/min, P>0.05; the second week (138.38±21.68) ml/min vs (143.05±20.69) ml/min, P>0.05]。 Three-four weeks after MCT injection, mean pulmonary artery pressure significantly increased in MCT group compared to control group [the third week(34.00±4.40)mm Hg vs(24.30±3.16)mm Hg, P<0.01; the forth week (37.30±3.53) mm Hg vs (23.70±3.65) mm Hg, P<0.001]. Cardiac output was significantly lower in PAH rats than in control rats [the third week (156.49±15.52) ml/min vs(156.49±15.52) ml/min, P>0.05; the forth week (138.38±21.68) ml/min vs (143.05±20.69) ml/min, P<0.001].[Conclusion] Pulmonary vascular resistance in rats could be reliably detected using the improved hand-made PE-50right heart catheter.Establishment of the rat models of pulmonary hypertension induced by monocrotaline is relatively simple, easy operation, and is an reliable animal model of pulmonary hypertension. Part Two Evaluation and analysis of rat models of chronic pulmonary hypertension in MRI[Objective] The aim of our study was to establish a rat model of chronic pulmonary hypertension and discuss the analysis and evaluation of the model in MRI.[Methods] Eighty Sprague-Dawley rats were randomly assigned into two groups:control group, and monocrotaline (MCT) group (60mg/kg) to establish the rat models of pulmonary hypertension In the process of pulmonary hypertension,Synchronously measured using the right heart catheter and MRI,to Assessment and analysis the change of Ventricular Function.[Results] MRI dynamic monitoring results showed that in the first two weeks, Pulmonary artery diameter between rats in PH and the control group showed no significant difference [the first week (3.05±0.19) mm vs (3.04±0.16)mm, P>0.05; the second week(3.05±0.23)mm vs (2.98±0.19) mm, P>0.05]; the right cardiac output between rats in PH and the control group was no significant difference [the first week (113.97±18.5) ml/min vs (103.44±10.92) ml/min, P>0.05; the second week (94.84±9.19) ml/min vs (92.50±10.04)ml/min, P>0.05]。Three-four weeks after MCT injection, the right cardiac output significantly reduced in MCT group compared to control group[the third week (72.83±11.73) ml/min vs (93.87±11.43) ml/min, P <0.05; the forth week (80.54±14.76) ml/min vs (96.74±10.75) ml/min, P<0.05]. The right ventricular end diastolic volume increased in MCT group compared to control group [the third week (0.44±0.04) ml vs (0.38±0.03) ml, P<0.05; the forth week (0.45±0.03) ml vs (0.38±0.03) ml, P<0.05].Pulmonary artery diameter showed no significant difference[the third week(3.07±0.19)mm vs(2.98±0.14)mm,P>0.05; the forth week(3.12±0.22) mm vs(3.10±0.18)mm, P>0.05]; Part of the ventricular septum with different degrees of change.[Conclusion] MRI can detect the rats model with chronic pulmonary hypertension, the right ventricular volume, cardiac output can be suggestive of pulmonary arterial hypertension, and ventricular septal curvature change also indirectly reflects the severity of pulmonary arterial hypertension.
