| Objective: To investigate Sufentanil on intestinal ischemia-reperfusion model of liver cellapoptosis and Bcl-2/Bax expression, and the role of opioid receptors in the process.Methods: 72 healthy adult Wistar rats, weight 200-250g, either male or female, wererandomly divided into 9 groups ( n = 8 ) : sham operation group ( group Sham), ischemicreperfusion group ( IR group), sufentanil preconditioning group (SPC group), group NTD ( deltaopioid receptor blocking agent group ), group CTOP ( mu opioid receptor blocking agent group ),group nor-BNI ( kappa opioid receptor blocking agent group), three kinds of opioid receptorantagonist intervention group ( NTD+SPC group, CTOP+SPC group, nor-BNI+SPC group). Byclamping the superior mesenteric artery (SMA) 1h, then the restoration of blood supply of 6hproduction of intestinal ischemia reperfusion injury model, after the end of the experiment willtake the left liver lobe making specimens. Light microscopy, transmission electron microscopicobservation of histological and ultra structural changes of liver. Using immunohistochemistryand MIAS-2000 on liver tissue Bcl-2, Bax protein expression were semi quantitative analysis,TDT labeling technique (TUNEL) method for the detection of liver cell apoptosis index (AI).Results:â‘ The result of light microscopy: compared with Sham group, IR group, threegroups of opioid receptor blocker controls liver tissue change obviously, SPC group, threegroups of opioid receptor antagonist intervention group than in IR group change loss.â‘¡Theapoptosis of liver cells (AI) changes: compared with group Sham, group AI was increased (P <0.05). Compared with IR group, SPC group of liver cell AI value decreased ( P < 0.05 ); NTDgroup, nor-BNI group, CTOP three group AI showed no significant differences ( P > 0.05 ).Compared with SPC group, NTD+SPC group, nor-BNI+SPC group, CTOP+SPC group of livercell AI were increased ( P < 0.05 ), AI value differences between the three groups had nostatistical significance ( P > 0.05 ).â‘¢The Bcl-2, Bax protein and Bcl-2/Bax ratio changes of OD:compared with group Sham, group Bcl-2, Bax expression were increased, Bcl-2/Bax ratiodecreased ( P < 0.05 ); compared with IR group, SPC group, Bcl-2 expression, Bcl-2/Bax ratioincreased, Bax expression ( P < 0.05 ) in NTD group, nor-BNI group, CTOP group, Bcl-2 Baxprotein content showed no significant differences ( P > 0.05 ). Compared with SPC group,NTD+SPC group, nor-BNI+SPC group, CTOP+SPC group, Bcl-2 expression, Bcl-2/Bax ratiodecreased, elevated Bax expression ( P < 0.05 ); between the three groups Bcl-2, Bax proteincontent had no significant difference ( P > 0.05 ).Conclusion:â‘ The intestinal ischemia reperfusion injury can be caused by a distant organliver cell apoptosis.â‘¡The sufentanil preconditioning can be adjusted by the Bcl-2, Bax proteinexpression, relieve liver cell apoptosis, on rat intestinal ischemia reperfusion of rat liver producesa protective effect.â‘¢The Delta, kappa, Mu three opioid receptor mediates the protective effect of sufentanil. |
