Fermentation has existed in Chinese history for thousands of years. After the discovery of the first fermentation drug in Han and Jin dynasties, fermentation methods became a choice of processing Chinese material media. In terms of the traditional fermentation method, solid fermentation is usually completed under natural condition with normal pressure and temperature, which means that there is no control of spawn, temperature, and humidity. In this case, it requires longer Incubation Period. Traditional fermented processing way though in slices plays an important role in the production, but its product quality as the change of environmental conditions and fluctuation, control is poorer. Modern Chinese medicine fermentation technology is adopted in the thousands of years of traditional fermentation, and on the basis of the strains on the conditions of fermentation and the control, can change the performance "of the directional drugs, or according to the characteristics of drugs from between the combination, using a single or mixed bacteria strains directional fermentation. The modern fermentation technology is used in Chinese medicine fermentation processing, which will be good for standard fermentation yinpian production process, improve the quality of traditional Chinese medicine fermentation processed products and the production efficiency. This essay focuses on using fixed fermentation with Rhizoma Arisaematis and bile to produce traditional Quj——Dan nanxin, aims to the fermentation process conditions, the chemical composition bile acid as the index, takes the best fermentation conditions for an investigation, discusses the fermentation process of bile acid material changes, preliminary explanation function mechanism of fermentation, For fermentation on traditional Chinese medicine study on the impact of the effective components provide reference. The study mainly includes the following several parts content:1. First established the qualitative and quantitative analysis method, in order to guide the optimal fermentation conditions.UV-VIS Spectrophotometry and HPLC-ELSD are used to establish the bile acid compounds in the determination method. Firstly, Glacial acetic acid and sulfuric acid UV-VIS Spectrophotometry was used to determined the content of cholic acid in Niudan powder. Because cholic acid is a weak acid on ultraviolet absorption and the sensitivity is not high, the method just can measure the total content of cholic acid. It can’t be used for multi-component quantitative analysis, so the measuring method switches to high performance liquid chromatography (HPLC). This paper mainly USES is evaporative light-scattering detector, mobile phase is Methanol-water-glacial acetic acid (80:20:0.05), drift tube temperature for80. This method chromatographic peak of the baseline stable, and in the sample reference substance peak of the same retention time of peak appearing. Reference substance is good linear relationship, simple and stable and repeatability, and get the LC-MS method provided the bile acid and deoxycholic acid two substances chromatographic peak. This method can be used to lignad in the gall acid and deoxycholic acid content is determined2. In establishing after analysis method, the fermentation process. First of all, according to the literature, fermentation processing change three fermentation factors, respectively is raw material, temperature and humidity, sampling for eight times, for the preparation of the product. Then we use the HPLC-ELSD method for the samples quantitative analysis. The experimental result obtained that the cholic acid content of1.8%and the deoxycholic acid content of0.73%in Niudan powder. The deoxycholic acid content ranges between3.04%to4.63%in Dan nanxing which cultured in the Niudan powder raw materials at37℃. The deoxycholic acid content ranges between1.9%to2.9%in Dan nanxing which cultured in the Niudan powder raw materials at26℃. The cholic acid content ranges between3.61%to7.42%in Dan nanxing which cultured in ox-bile raw materials at37℃. The cholic acid content ranges between1.2%to4.7%in Dan nanxing which cultured in ox-bile raw materials at26℃. But we can’t detected deoxycholic acid exists. From the result, we can come to NiuDan powder as raw materials, in37℃dry fermentation can get bile acid and deoxycholic acid content of the highest. The experimental results can be the method based on used HPLC-ELSD for Niudan powder and Dan nanxing quantitative analysis.3. The Dan nanxing of the sold products as research vectors, strains of fermentation preliminary inquiry. The erperimental results show that it is the bacillus in Dan nanxing samples. For specific identification of strains, still need further investigation.The above studies have shown that, HPLC-ELSD can be used as cholic acids compounds qualitative and quantitative analysis method. In the37℃dry conditions, it can get with a high content of acid compounds bravery lignad fermentation product, which can be used as a lignad fermentation technology of reference.
