Cancer is a complex and multi-stage disease, which can be leaded by mutationof genes, gain or loss of genetic materials (copy number variation), microRNA ordisruption of the epigenome. DNA methylation is the most important form ofepigenetic,and aberrant DNA methylation plays a vital role in the cancer process.With the development of high-throughput technologies, DNA methylation chip hasbeen widely used in cancer research. Abnormal DNA methylation to identify cancergenes can not only help the understanding of cancer biology mechanism, and thediagnosis and treatment of cancer. In past research on biological markers, biologicalmarkers found in the different experiments of the same cancer type overlap isusually very low, as the signs of the disease in the use of DNA methylation needs ofits repeatability full evaluation. The work of genetic markers do not overlap, butthey may affect the same biological function. Therefore, in the evaluation ofdifferentially methylated genes repeatability, we should consider consistency ratherthan the surface of the genes in the biological function of the proportion ofoverlapping genes. In addition, studies have found that the level of methylation incancer is independent of each process is not clear, but the abnormally low biologicalfunctions of methylation of the gene promoter region, so we separate the function ofthe level of methylation differences.Firstly, from the sites or genes two levels, the differentially methylated genesfound in the two sets of colon cancer methylation data found that the methylation markshadn’t high repeatability, but they have a high degree of the same change direction;Next, we proceed from the functional designed differentially methylated gene list offunctions repeatability index POE value evaluation repeatability, found differentiallymethylated genes with a high degree of functionality can be repetitive, the resultssuggest that the functional level assessment consistency of differentially methylatedgenes are reasonable; Finally, by comparing the level of enrichment of methylatedgenes function, we found that hypermethylation of genes affect the development process, and hypomethylation of gene mainly affects the immune andstress responses.
